芦沙坦在压力超负荷心肌肥大作用的形态计量学分析

摘  要:目的观察压力超负荷心肌肥大大鼠的体视学变化,探讨血管紧张素受体拮抗剂芦沙坦(Los)在压力超负荷心肌肥大反应过程中的作用。方法建立腹主动脉缩窄性高血压大鼠模型,测定大鼠平均动脉压(MAP)、大鼠心肌的表面积密度(SV)、体积密度(VV)、比表面(S/V)及平均自由程(λ)等各项体视学参数并计算。结果COA大鼠的MAP明显增高,心肌横断面积及平均直径明显增大,SV、VV、S/V及λ等与对照组相比均有显著性差异;COA+Los组的MAP值大致恢复至正常水平,心肌横断面积、平均直径及SV、、VV等明显小于COA组,S/V及λ则增大。结论Los可明显改变体视学各项参数,对压力超负荷引起的高血压和心肌肥大具有明显逆转的作用。Objective To analyze the role of angiotension Ⅱ type 1 receptor antagionist losartan(Los) in the myocardial hypertrophy rats caused by pressure overload during investigating the change of stereological data. Methods The coarctation of the abdominal aorta (COA) hypertension model was established in Sprague-Dawley rats. The mean arterial pressure (MAP) were measured in each rat. the stereological parameters such as the myocardial surface density,the myocardial volume density, the myocardial specific surface and the mean free path etc. were measured and numerated. Results The MAP of COA rats was significantly increased, the myocardial traverse area increased and the average diameter, surface density, the volume density, the specific surface and the mean free path showed remarkable differences as compared with those in control group; the MAP of COA and losartan (Los) rats approximately returned to normal level and the myocardial traverse area, the average diameter, the surface density and the volume density were remarkable smaller than those in COA group; while the specific surface and the mean free path were increased. Conclu- sion Los can obviously change every stereological parameters and possess the action in normalizing the hypertension and reverse the myocardial hypertrophy caused by overloaded pressure.
