冠心苏合制剂中马兜铃酸A的含量

摘　　要：目的：建立马兜铃酸的含量测定方法，并测定不同厂家冠心苏合制剂中马兜铃酸A的含量，用于指导临床用药。方法：用高效液相色谱法进行分析，色谱柱为Alltech C18，流动相甲醇-水-醋酸（68：32：1），流速1．0mL·min^-1，柱温35℃，检测波长390nm。成药用甲醇-10％甲酸水提取。结果：马兜铃酸A在0．119～1．89μg，有良好的线性关系；平均加样回收率为99．0％，RSD0．63％。不同厂家冠心苏合丸（胶囊、滴丸）中，以北京同仁堂股份有限公司同仁堂制药厂（滴丸）中马兜铃酸A的含量最低（0．148mg·g^-1）；以三九企业集团（胶囊）中马兜铃酸A的含量最高（0．516mg·g^-1）。结论：本方法简便，准确，可用来测定冠心苏合制剂中马兜铃酸A的含量测定。Objective： To establish a determination method of aristolochic acid A in Guanxisuhe preparations by RP-HPLC. Method： The instrument used was Hewlett-Packard 1100 HPLC with a Alltech C18 column（4.6 mm× 250 mm, 5μm） .The mobile phase was methanol- water-acetic acid（68：32：1 ） and the flow rate was 1.0 mL·min^-1. The UV detection wavelength was 390 nm and the column temperature was at 35℃. The extracted solvent for the preparations was methanol solution contained 10% formic acid. Result： The calibration curve was linear （ r = 0.999 9） within the range of 0.119-1.89 μg for aristolochic acid A. The average recovery 99.0%, RSD 0.63% .Conclusion： The method with good linear relationship was convenient, quick, accurate, and suitable for the quality control of the aristolochic acid A in Guanxinsuhe and other traditional Chinese medicines containing aristolochic acid A.
