摘  要:为研究鸽新城疫病毒(PNDV)和鸽痘病毒(PPV)的血凝特性,同时为2种病毒的血凝试验(HA试验)提出技术参数,分别采用鸡和鸽红血细胞(RBC),在不同反应介质和条件下进行病毒血凝试验。结果表明:①PNDV既能凝集鸡RBC,又能凝集鸽RBC,而PPV只凝集鸡RBC;②PNDV对0.5%鸡RBC血凝价比对1%鸡RBC血凝价高1~2个稀释度,而PPV对0.5%鸡RBC血凝价比对1%鸡RBC血凝价高5个稀释度;③PNDV和PPV对鸡RBC的HA凝集强度呈线性变化,而PNDV对鸽RBC的HA凝集强度墨非线性变化;④进行PNDV和PPV的HA试验的理想技术参数为:用0.5%鸡RBC,以0.89%生理盐水或pH7.4PBS为反应介质,PNDV于37℃作用30min时转至室温观察,重复观察应于35min内结束,而PPV应于37C作用30min观察,观察后迅速放回37℃,重复观察应于10min内结束。To research on the hemocoagulation (HA) properties of PNDV and PPV and search for technical parameters for HA, red blood cells (RBC) of cocks and pigeons were adopted for HA of the viruses in different media and conditions. The results showed that :①PNDV can coagulate the RBC of cock and pigeon, while PPV only coagulates RBC of cock; ②The HA valence of PNDV in 0.5 % cock RBC is higher than that in 1% cock RBC by 1~2 dilution grades, and HA valence of PPV in 0.5% cock RBC is higher than that in 1% cock RBC by 5 dilution grades ; ③the HA percentage of PNDV and PPV with cock RBC demonstrated an linear trend as the virus is successively diluted but that of PPV with cock RBC demonstrated an non-linear pattern and ;④ the ideal technical parameters of HA with PNDV and PPV include 0.5% cock RBC, 0.89% NaCl or pH 7.4 PBS as medium, observation after 30 min of reaction of PNDV with the RBC at 37℃ and followed by necessary re-observation within 35 min at room temperature, and observation after 30 min of reaction of PPV with the RBC at 37℃ and followed by necessary re-cuhure within 10 min at 37℃ for observation.
