摘 要:为建立HPLC法测定小鼠、大鼠脑内谷氨酸、天冬氨酸、甘氨酸及γ-氨基丁酸的浓度,为进一步研究药物对脑内氨基酸含量影响提供测定方法.采用高效液相色谱法,色谱柱为Hypersil C8柱(150mm,4.6mm);流动相A液为0.14mol/L乙酸钠液:乙腈=470:30,B液为50%乙腈;流速1.5mL/min;柱温40℃;检测波长360nm;进样量20uL;梯度洗脱:B液于0~10min由18%~35%;10~17min由35%~80%;17~18min由80%~18%.结果显示,4种氨基酸在1.2~36ug/mL范围内呈良好线性关系(r值均大于0.9990).方法回收率均在90%~115%,日内RSD均小于5%,日间RSD均小于10%.结果表明,新建立的高效液相色谱法简便快速,准确,灵敏度高,适用于药物对小鼠、大鼠脑内氨基酸含量变化的研究.To establish an HPLC method to determine the content of glutate,aspartate,glycine and γ-GA. BA in mice and rat brain. The sample was separated on an Hypersil C8 column(150mm,4.6mm), A mobile phase: 0.14mol/L sodium 470mL,plus acrylonitrile 30mL, B mobile phase: 50% acrylonitrile (flow rate 1.0mL/min), column temperature was 470℃ and detection wavelength was 360nm, Gradient elution using A and B solvent system. Calibration curves were good linear within the concentration range of 1.2 - 36μg/mL for four kinds of amino acids. Recoveries of assay were 90%-115% and relative standard deviations for inter-day and intra-days were all less than 5.0% and 10% respectively. The HPLC method is simple, rapid, sensitive and accurate, It can be used for the contents study of amino acids in mice and rat brain.
