益气活血中药对缺血心肌降钙素基因相关肽免疫反应神经纤维的影响  

Effect of Yiqi Huoxue compound Chinese herb on calcitonin gene-related peptide immunoreactive fibers of ischemic myocardium

摘　　要：目的:观察益气活血中药复方制剂芪丹通脉片对心肌缺血大鼠的保护作用是否与增加降钙素基因相关肽免疫反应阳性神经纤维数量有关。方法:①实验于2004-11/2005-05在解放军第四军医大学神经生物实验室完成。选用清洁级SD雄性大鼠60只。随机将大鼠分为6组:空白对照组、模型组、阳性对照组、芪丹通脉片低剂量组、芪丹通脉片中剂量组、芪丹通脉片高剂量组,每组10只。②灌胃前依据大鼠体质量计算芪丹通脉干粉(由黄芪、丹参、当归、红花、桂枝等组成)低、中、高剂量组及阳性对照组给药量[分别为芪丹通脉干粉0.36,1.08,3.24g/(kg·d),单硝酸异山梨酯片15mg/(kg·d)],用蒸馏水配置成等体积混悬液。按10mL/(kg·d)每天灌胃1次,连续15d,空白对照组与模型组灌胃等体积生理盐水,第15天各组最后一次灌胃30min后,先连续观察并记录一段Ⅱ导的正常心电图,然后模型组及给药组分别腹腔注射垂体后叶素20U/kg造成心肌缺血模型,空白对照组腹腔内注射等剂量生理盐水,造模后继续监测心电图变化。在200倍光镜下观察心肌组织形态学改变(苏木精-伊红染色)。③对心肌中降钙素基因相关肽神经纤维行免疫组化染色,每一标本随机取3张切片,每组10个标本观察30张切片,150个视野,以20倍镜下视野为单位,对随机取出的切片进行观察。将心肌中降钙素基因相关肽免疫反应阳性神经纤维的密度,按每20×光镜视野下阳性纤维数定为如下3个等级:Ⅰ级:0～5根阳性纤维;Ⅱ级:5～10根阳性纤维;Ⅲ级:10根以上或呈网状。④等级资料差异性比较采用秩和检验。结果:大鼠60只均进入结果分析。①模型组有明显心肌缺血病理改变。芪丹通脉片各剂量组心肌缺血病理改变均有不同程度改善。②大鼠腹腔注射垂体后叶素后心电图表现为心肌缺血变化。模型组降钙素基因相关肽免疫反应阳性神经纤维数AIM： To investigate whether the protective effect of Yiqi Huoxue compound Chinese herb, Qidantongmai tablets （QDTMT） on myocardial ishemia in rats is related to the increasing quantity of calcitonin generelated peptide positive immunoreactive nerve fibers. METHODS： （1）The experiment was performed in the Department of Neurobiology of the Fourth Military Medical University of Chinese PLA from November 2004 to May 2005. Sixty clean-grade SD male rats were selected and randomly divided into six groups： blank control group, model group, positive control group, QDTMT low dosage group, QDTMT medium dosage group and QDTMT high dosage group with 10 rats in each group. （2）According to body mass of rats, the required dosages of QDTMT, which was made up of membranous milkvetch root, dan-shen root, Chinese angelica, safflower and cassia twig, etc., was calculated in low, medium, high QDTMT dosage groups before gastric perfusion [0.36, 1.08, 3.24 g/kg per day, respectively and isosorbide mononitrate 15 mg/kg per day]. The medicines were dissolved in distilled water to get equal volume solutions for each group. 10 mL/kg solution was given pet day with gastric perfusion, once a day for 15 days. Same volume saline was used as placebo with gastric perfusion in the blank control group and model group. At the 15^th day, 30 minutes after the last infusion was executed, and electrocardiogram was observed and recorded, subsequently the rat experimental acute myocardial ischemia model was established by 20 U/kg pituitrin with jntraperitoneal injection in medicine treated groups and model group. At the same time, same volume saline was used as placebo with intrapefitoneal injection in the blank control group. After the model was successfully established, the electrocardiogram was observed and recorded again.
