摘  要:目的:观察益气化瘀利水方对实验性兔骨关节炎软骨中转化生长因子β1的影响。方法:实验于2003-07/12在上海中医药大学脊柱病研究所完成。取雄性6月龄新西兰大白兔20只,随机分为正常组、模型组、益气化瘀利水方组、氨糖美辛组。每组5只。按Colombo法建立家兔膝骨关节炎模型,术后第5~8周给药。益气化瘀利水方组给予益气化瘀利水方0.09g/(kg·d),氨糖美辛组给予氨糖美辛0.09g/(kg·d),正常组及模型组均未给药。各组于术后第9周取材,取右胫骨平台关节软骨,采用免疫组织化学方法进行染色,光镜下观察,并利用图象分析测定其转化生长因子β1表达的积分吸光度。结果:20只白兔均进入结果分析。①各组关节软骨光镜观察结果:模型组和氨糖美辛组,关节软骨损伤严重,转化生长因子β1主要在发生簇集的软骨细胞胞膜和胞浆内表达,益气化瘀利水方组软骨损伤较氨糖美辛组明显减轻。②各组关节软骨切片转化生长因子β1染色强度的积分吸光度图像分析:正常组与益气化瘀利水方组无明显差异(6.64±1.59,5.49±3.09,P>0.05);模型组与氨糖美辛组无明显差异(23.16±9.38,20.09±9.17,P>0.05);模型组较正常组和益气化瘀利水方组明显增高(F=8.96,P<0.01)。结论:益气化瘀利水方可抑制骨质增生,其作用机制之一可能是通过调控转化生长因子β1而达到的。AIM: To observe the effect of Yiqi Huayu Lishui prescription on transforming growth factor-beta 1 (TGF-β1) in cartilage in osteoarthritis rabbits. METHODS: The experiment was conducted at the Institute of Spondylopathy, Shanghai University of Traditional Chinese Medicine from July to December 2003. Twenty male New Zealand rabbits aged 6 months were selected and randomly assigned into normal group, model group, Yiqi Huayu Lishui prescription group (Chinese herb group) and Glucosamine Indometacin Entric-coated Tablets group (NSAIDs group) with 5 rabbits in each group. Osteoarthritic models were made according to Colombo method. The drug was given at 5-8 weeks. The rabbits in the Chinese herb group were given with 0.09 g/kg per day Yiqi Huayu Lishui prescription group. Those in the NSAIDs group were treated with 0.09 g/kg per day NSAIDs. Those in the normal group and model group were not treated with any drug. Materials were drawn at the 9^th week after operation in every group. Articular cartilage of right tibia was gained to stain with immunohistoehemical method and observed under the light microscope. Integral absorbance (A) of the expression of TGF-β1 was detected with image analysis. RESULTS: Totally 20 rabbits were involved in the result analysis. (1) Observational result of light microscope on articular cartilage of every group: The articular cartilage injured severely in the model group and the NSAIDs group. TGF-β expressed chiefly in membrane and plasma of cartilage cell with cluster. The injury of articular cartilage relieved significantly in the Chinese herb group as compared with the NSAIDs group. (2) Image analysis of A of staining intensity of TGF-β1 of model in cartiIage of every group: There was no obvious difference between normal group and Chinese herb group (6.64±1.59,5.49±3.09,P 〉 0.05).
