免疫印渍试验在斯氏肺吸虫病血清学诊断中的意义

摘  要:应用免疫印渍试验(Immunoblottingtest)分析斯氏肺吸虫成虫、幼虫和囊蚴期的虫体可溶性抗原.结果显示感染斯氏肺吸虫的人和动物血清可识别22、24和26KD抗原蛋白带.依据病人血清对上述抗原组仿的识别,用免疫印渍试验诊断24例斯氏肺吸虫病人,皆为阳性,而对照组显阴性.与Dot-ELISA比较试验的敏感性和特异性,两试验的阳性检出事无显著性差异(P>0.05).Dot-ELISA的交叉反应率为18.75%(6/32),而免疫印渍试验无交叉反应.表明斯氏肺吸虫的22、24和26KD蛋白带为特异性诊断抗原,免疫印渍试验为斯氏肺吸虫病的高度敏感和特异的血清学诊断方法.The soluble antigens of metacercariae, larvae and adult worms of Pagumogonimus skrjabini were analysed by using immunoblotting. 22, 24 and 26KD protein band of adult fluk were recognized in sera of humans and animals that were infected with P. skrjabini. Based 0n the results humans mentioned above, 24 cases of pagumogonimiasis diagnozed with this methed each showed pos itive reaction in recognition of 22 t 24 and 26KD bands whereas the control sera showed negative reac-tion. In comparison with Dot-ELISA for test sensitivty and speificity, X2 test revealed that there was no striking difference between the positive rates of the two metheds. No cross reaction occured with immunoblotting while 18. 7% (6/32) of the sera were positive in the Dot-ELISA test. It is thus con-cluded that 22, 24 and 26 KD antigenic proteins of P. skrjabini are useful diagnostic antigens. Im-munoblotting is considered an effective specific and sensitive serediagnostic method for pagumogo-nimiasis.
