黄角颗粒对MCAO大鼠神经细胞bc1-2、bax表达影响的实验研究

摘　　要：目的研究黄角颗粒对大鼠大脑中动脉栓塞(MCAO)模型脑神经细胞bc1-2、bax表达的影响。方法将MCAO大鼠分为中药大、中、小剂量组,西药组及模型组,另设正常组作对照;观察各组大鼠脑神经细胞bc1-2、bax表达的变化。结果模型组大鼠脑组织bc1-2、bax阳性细胞数显著高于正常组,bc1-2/bax比值显著低于正常组;中药大、中、小剂量组及西药组脑组织bax阳性细胞数显著低于模型组,bc1-2及bc1-2/bax比值显著高于模型组;中药大剂量组脑组织bc1-2、bax阳性细胞数与西药组相近,其bc1-2/bax比值显著高于西药组。结论黄角颗粒能下调MCAO大鼠脑组织促凋亡基因bax水平,上调抗凋亡基因bc1-2水平,提高bc1-2/bax比值;此可能是其抗急性缺血性神经细胞凋亡的机制之一。Objective:To observe the effect of Huangjiao Granule(黄角颗粒) on the Expression of bcl-2,bax in neuron of MCAO rats. Methods:Choosing 60 old health Wister rats,to divide them into 6 groups at random:Huangjiao Granule small dose group,Huangjiao Granule middle dose group,Huangjiao Granule large dose group,medicine group,model group,normal group. Then observe the expression level of neuronal cells bcl-2,bax in the 6 groups. Results:The number of brain tissue positive cells of bcl-2,bax in the model group were significantly higher than that in the normal group,bcl-2/bax level was significantly lower than that in the normal group.The number of brain tissue positive cells of bax of Huangjiao Granule small dose group,of Huangjiao Granule middle dose group and of Huangjiao Granule large dose group,of medicine group were significantly lower than that of the model group,the number of brain tissue positive cells of bcl-2 and bcl-2/bax of those group were significantly higher then that of the model group.The number of brain tissue positive cells of bcl-2,bax in Huangjiao Granule large dose group was similar to medicine group,the bcl-2/bax was significantly higher then that of the medicine group. Conclusions:Huangjiao Granule can cut down the levels of the brain tissue apoptosis gene bax of MCAO rats,raise the level of antiapoptosis gene bcl-2,raise the level of bcl-2/bax.Perhaps this is the reason that Huangjiso Granule can cut down the levels of the MCAO Rat acute cerebral ischemia apoptosis gene.
