摘 要:目的探讨川芎嗪在TNF-α蛋白表达及大鼠腹膜粘连形成中的作用。方法采用盲肠刮擦及切除右侧壁腹膜和腹膜下一层肌肉的方法制作S-D大鼠腹膜粘连模型;关腹前注射川芎嗪注射液(约为50mg/kg)和生理盐水;术后7d行二次开腹手术,取粘连组织,光镜下观察粘连组织病理变化。链霉亲和素免疫组化法(SABC法)检测粘连组织中TNF-α蛋白的表达量。放免法检测血液中TNF-α的浓度。结果川芎嗪组粘连面积明显减少,川芎嗪组粘连评分结果(2.10±0.99)明显低于模型组(3.80±0.42)和生理盐水组(3.60±0.70)(P<0.05),模型组和生理盐水组的TNF-α呈较一致的阳性反应,川芎嗪组的TNF-α呈现较弱阳性的反应或不表达,其染色强度评分(0.08±0.14)明显低于模型组(1.39±0.61)和生理盐水组(1.32±0.55)(P<0.05)。血液中的TNF-α浓度值在模型组(2.24±0.47)、生理盐水组(2.58±0.38)和川芎嗪组(2.51±0.42)三组之间差异无显著性(P>0.05)。结论川芎嗪通过抑制粘连组织中TNF-α的表达来减低大鼠腹膜粘连的程度,但对血液中的TNF-α含量无影响。【Objective】 To investigate the role of Ligustrazine in the expression of TNF-α and adhesion formation in rats. 【Methods】 The uniform defects on the peritoneal wall and cecal surface were developed to establish the rate adhesion model. Prior to complete closure, Ligustrazine (50 mg/kg) or 0.6 mL normal saline was instilled into the abdominal cavity of the Ligustrazine group or the normal saline group respectively. All rats were killed 7 days later. The scorns of adhesions area were measured. The immunohistochemical technique (Strept-Avidin-Biotin-enzyme Complex, SABC) was used to identify the volume of TNF-α. The radioimmunity technique was used to identify the coucentration of TNF-α in serum. 【Results】 The seores of intraperitoneal adhesion area was signifieantly lower in the Ligustrazine group than in model group and the normal saline group (2.10±0.99) versus (3.80±0.42), (3.60±0.70); P〈0.05. There was no significant difference in the concentration of TNF-α in serum among group L, group M and group N rats (2.51±0.42), (2.24±0.47) and (2.58±0.38); P〉0.05. The immunohistochemical staining showed positive expressions of TNF-α in group M and group N rats, but slightly posilive expressions or no expressions in group L. The staining was situated in the maerophage cytoplasm in the adhesion tissue. The scores of staining intensity was significantly lower in group g rats than in group M and group N (0.08±0.14) vinous (1.39±0.61), (1.32±0.55); P 〈 0.05. There was no significant difference in the staining intensity between group M and group N rats. 【Conclusions】 The Ligustrazinc can reduce the area of postoperative intraperitoncal adhesions by restraining restrain the expressions of TNF-α in adhesion tissues, but has not influence on TNF-α in serum.
