摘 要:目的探讨镉对人胚肾细胞(HEK239T)凋亡和血红素氧合酶-1(HO-1)表达的影响。方法 MTT法测定细胞增殖活性,流式细胞仪检测镉诱导人胚肾细胞凋亡,应用逆转录多聚酶链反应 (RT-PCR)及Western免疫印迹法检测镉对人胚肾细胞HO-1 mRNA及蛋白表达的影响。结果 5.0、 10.0、20.0、40.0μmol/L CdCl2诱导人胚肾细胞后,凋亡率分别为11.90%±0.28%、9.27%±1.73%、 9.79%±0.67%、8.97%±1.60%,均明显高于对照组(6.69%±0.46%),差异有统计学意义(P< 0.01)。10~40μmol/L CdCl2均可高度诱导人胚肾细胞HO-1表达,随着剂量增高缓慢上升并达到平台期,随时间的延长略有降低。结论镉可诱导人胚肾细胞凋亡,上调HO-1的表达。Objective To investigate apoptosis and expression of heine oxygenase-1 (HO-1) induced by cadmium in human embryonie, kidney cells ( HEK239T). Methods The MTT method was used for determining the cell proliferation activity. The apoptosis was determined by the flow eytometry. The HO-1 mRNA expression and protein level were detected by RT-PCR method and Western blot respectively. Results The ratios of apoptosis in HEK239T ceils were 11.90% ±0,28% .9.27% ± 1.73% ,9.79% ± 0.67% and 8.97% ± 1.60% at the concentration of 5.0,10.0,20.0 and 40.0μmol/L CdCl2 respectively, higher than those in the control group (6.69 % ± 0.46% ) with the significant difference ( P 〈 0.01 ). The CdCl2 of between 10 and 40 μmol/lL could highly induce the expression of HO-1 of the human embryonic kidney cells. The expression would increase slowly till the flat stage with the increase of the dosage and then would decrease slightly over time, Conclusion The cadmium can induce the apoptosis of the human cmbryonic kidney cells and up-regulate the expression of HO-1.
