摘 要:目的研究亚硒酸钠对氯化镉诱导的大鼠肝脏端粒酶逆转录酶(TERT)mRNA、c-Myc蛋白、p53蛋白表达的影响。方法健康雄性SD大鼠随机分为6组,对照组、硒组(5 μmol/kg亚硒酸钠)、 5 μmol/kg氯化镉组、10μmol/kg氯化镉组、硒(5μmol/kg亚硒酸钠)+5 μβmol/kg氯化镉组、硒(5 μmol/kg 亚硒酸钠)+10 μmol/kg氯化镉组。每组动物5只,染毒48 h后取出肝脏,用RT-PCR方法检测TERP 基因的表达,用免疫组织化学方法检测c-Myc蛋白和p53蛋白的表达。结果与对照组比较,5μmol/L kg氯化镉组和10μmol/kg氯化镉组TERP表达增多,10 μmol/L kg氯化镉组c-Myc蛋白表达增多,5 μmol/Lkg氯化镉组和10μmol/L kg氯化镉组p53蛋白表达增多。TERT的表达,硒+10/μmol/L kg氯化镉组与10 μmol/L kg氯化镉组比较降低;硒+10 μmol/Lkg氯化镉组比10μmol/Lkg氯化镉组c-Myc蛋白表达降低;硒+5 μmol/Lkg氯化镉组、硒+10 μmol/L kg氯化镉组与相对应的镉组比较p53蛋白表达降低。结论镉在5~10 μmol/L kg的剂量条件下可以诱导大鼠肝脏高表达TERT、c-Myc蛋白和p53蛋白,而硒对镉引起的TERT、c-Myc蛋白和p53蛋白的表达增高具有一定的拮抗作用。Objective To study the effects of sodium selenite on expression of telomerase reverse transcriptase mRNA, c-Myc and p.53 induced by cadmium chloride in rat liver. Methods Male SD rats were divided randomly into 6 groups,each group had 5 animals. The groups comprised the control group, Se group (5μmol/kg sodium selenite),5μmol/kg cadmium chloride group, 10μmol/kg cadmium chloride group, Se (5 μmol/kg sodium selenite) + 5 μmnol/kg cadmium chloride goup,Se (5μmol/kg sodium selenite) + 10μmol/kg cadmium chloride group. After 48 hours of the first injection, the expression of TERT mRNA was measured with RT-PCR and c-Myc, and p53 proteins were measured by immunohistochemistry method. Results Compared with control group, the expression of TERT was increased in 5μmol/kg Cd goup and 10μmol/kg Cd group, c-Myc protein was increased in 10μmol/kg Cd group, and the expression of 1:63 protein was increased in 5 μmol/kg group and 10 μmol/kg Cd group. TERT expression in Se + 10μmol/kg Cd group was lower than that of 10μmol/kg Cd group significantly, c-Myc protein was decreased in Se + 10μmol/kg Cd group compared with 10 μ/kg Cd group.p53 protein of Se + 5μmol/kg Cd group and Se + 10μmol/kg Cd group were decreased significantly compared with 5 μmlol/kg Cd group and 10 μmol/kg Cd group respectively. Conclusion The cadmium at the doses of between 5 and 10 μmol/kg can activate TERT and up-regulate c-Mye and p53 proteins. The selenium at the dose of 5 μmol/kg has the antagonistic effect on expression of TERT, c-Myc and p53 induced by cadmium in rat liver.
