摘 要:目的:探讨黄芪当归合剂对实验性缺血性肾损伤大鼠肾小管上皮细胞凋亡的保护作用。方法:建立大鼠实验性缺血性急性肾损伤模型,在缺血30 m in再灌注不同时间点检测血尿素氮(BUN)、血肌酐(SC r)和尿液中N乙酰βD氨基葡萄糖酐酶(NAG)。同时取肾组织,苏木素伊红(HE)染色,光镜下观察细胞形态学变化。免疫组织化学方法检测B cl 2和B ax基因的蛋白表达,观察黄芪当归合剂对上述表达的影响。结果:缺血性急性肾损伤时肾脏存在明显的细胞凋亡;B cl 2和B ax基因的蛋白表达主要在近曲肾小管和远曲肾小管,肾小球很少;肾脏缺血性损伤时B cl 2基因表达增加,B ax基因表达中量增加,B cl 2/B ax比率升高。与缺血/再灌注组比较,黄芪当归合剂组B ax表达明显增加,B cl 2/B ax比率降低,肾脏损伤亦减轻。结论:黄芪当归合剂对急性肾损伤具有显著的保护作用,其机制可能与其降低B cl 2/B ax比率有关。Objective: To investigate the protective effects of astragalus -angelica mixture (黄芪当归合剂) on apoptosis of renal tubular epithelium in rats with experimental ischemic acute renal injury. Methods: Experimental acute ischemic renal injury model of rats was induced. Levels of blood urea nitrogen (BUN), serum creatinine (Cr) and N -acetyl amino glucosidase (NAG) in urine were detected at different time points of ischemia for 30 minutes and reperfusion. The cell morphologic changes of renal tissue were observed by hematoxylin and eosin (HE) staining. Expressions of Bcl - 2 protein and Bax protein were detected by immunohistochemistry and the effect of astragalus - angelica mixture on it was observed. Results : Cell apoptosis was obvious in kidney in acute ischemic renal injury. Bcl - 2 and Bax were mainly expressed in proximal and distal convoluted renal tubules, and much less expression in kidney glomerulus. The expressions of Bcl - 2 mRNA, Bax mRNA and Bcl - 2/Bax ratio increased in the kidney with acute ischemic injury. Compared to the ischemia/reperfusion injury group, the expression of Bax mRNA increased, Bcl -2/Bax ratio decreased, and the renal pathological damage was ameliorated in the group treated by astragalus -angelica mixture. Conclusion: Astragalus -angelica mixture has protective effects on injuried kidney, the mechanisms may be related to the decrease of Bcl - 2/Bax ratio.
