油菜无菌苗最佳生长条件的探讨

摘　　要：以甘蓝型油幕种子作为试验材料，对无菌苗培养前的种子消毒荆种类厦不同消毒时间、浓度等的消毒效果进行了比较研究；对影响无菌苗生长的培养基配方、pH值厦生长时间等因素进行了探讨。结果表明：油幕无菌苗的最佳生长条件为：70％酒精表面消毒1～2min后，用15％H202消毒35min，无菌水冲洗3～5次，无菌水浸泡种4～15h，接种至1／2MS培养基＋20g／L蔗糖＋7．5g／L琼脂，pH值为5．8，25℃，生长4～6d即可作为转基因用无菌苗。The experiment was conducted to investigate effects of different disinfectant agents, disinfectant times and concentrations on the rapeseed before cultured and effects of medium, pH and growing period on growth of the sterile seedling. The results showed that the aseptic rape seedling for transgenic could be obtained throughout the rapeseed was sterilized 1-2 rain with 70% alcohol, sterilized 35 min with 15% H2O2, washed 3-5 times with aseptic water and soaked in aseptic water 4-15 h, and then was cultured for 4-6 d on 1/2MS ＋20g/L sugar＋7.5 g/L agar with pHS. 8 under 25℃.
