摘  要:分离蛋白质的梯度洗脱色谱中,当洗脱液B液达到一定的百分浓度前,可以多次累加进样,保留值及分离度无可觉察的变化。蛋白质的分析和制备时,应用此方法可以大大增加柱子的处理量,提高检测灵敏度,节省时间和消耗。An abrupt change of the elution curve of proteins in the gradient elution chromatography was observed. When the composition of mobile phase is far away from the position of the abrupt change, the multiple injection of proteins before and after starting a gradient elution for one chromatographic separation in the IEC, HIC and RPC can be performed. Thus the variation of protein retention time and capacity factor are not significant. The two operation processes, concentration and separation, can be finished on one chromatographic procedure. This method improves the analytical sensitivity. Also a larger size sample can be injected by many times in one chromatographic run to save time and decrease the consumption.
