摘 要:目的:考察3种不同相对分子质量的聚乙烯毗咯烷酮(PVP)包覆的β-榄香烯脂质体的包封率,确定最终处方。方法:采用正交设计优化处方与制备工艺;采用乙醇注入法制备不同相对分子质量PVP包覆的β-揽香烯脂质体;采用微柱离心-气相色谱法测定脂质体包封率。结果:正交试验结果证明利用1%PVP-k30包覆,载药量为0.5%,磷脂用量为6%,磷脂与胆固醇质量比例为6:1为最佳处方。结论:脂质体用PVP包覆后包封率为(97.19±0.27)%,与普通脂质体相比未见明显变化,粒径则略有增加。Objective: To assess the inclusion rate of β-elemene liposomes complexes with 3 kinds of polyvinylpyrrolidones (PVPs). Methods:The formulation and preparation were optimized by an orthogonal test. The liposomes inclusion complexes were made using an ethanol injection method, The inclusion rate was measured by a minicolumn centrifugation-gas chromatography method. Results:The optimal formulation was composed of 1% PVP-k30, 0.5%β-elemene, 6% phospholipids and 1% cholesterol. The inclusion rate was (97.19 ±0.27)% , which showed no significant difference from conventional liposomes except a slight increase of particle diameter. Conclusion:The PVP-complexed β-elemene liposomes were attained.
