摘  要:目的观察抗癌灵对小鼠的急性毒性反应,为拟定临床安全剂量提供依据。方法采用急性毒性实验方法,以最大浓度最大体积给小鼠灌胃给药,连续观察小鼠行为活动等指标和毒性反应程度,并在实验结束时处死小鼠进行剖检,以获得初步毒性资料。结果7d内各组小鼠均健康存活,未见明显中毒症状,最大耐受量相当于临床人用剂量的360倍。药物对小鼠的体重增长和食物利用率无不良影响;脏器系数除给药组雄性小鼠的肾脏外,其他心、肝、脾、肺、胸腺、睾丸、卵巢等均无不良影响;各组的血象及血液生化指标,与对照组相比较,无显著差异;肉眼观察解剖的脏器,各标本表面及切面未见异常改变,各脏器病理检查无异常。结论抗癌灵未见急性毒副作用,临床应用安全。Objective To study acute toxicity of the drug anti-tumor agent on mice, thus offering evidence for clinical safe dose. Methods An acute toxicity test was conducted. Anti-tumor agent in the highest concentration and largest amount was given to the mice by stomach feeding, and behavioral indexes and toxicity degree were continuously observed. Then in the end of the test, the mice were executed in order to get primary toxicity materials. Results All mice healthily survived during the period of test, and no obvious symptoms were observed. The maximum tolerance dose was as 360 times as that used in clinic. The drug had no harmful effect on body weight, food utilization as well as organ coefficieats except that of male mice's kidney. The hemogram and blood chemical indexes of the grOupS were not significantly different from those of control group, Observed by naked eyes, there were no abnormal changes on surfaces and transects of organs. The results of pathological examination on all organs were also normal, Conclusions No acute toxicity is found with anti-tumor agent, and it may be safe to be used in clinic.
