摘  要:目的探讨丹参素(DSS)保护肝损伤的作用及其可能机制。方法用硫代乙酰胺(TAA)构建急性重型肝损伤大鼠模型。24只雄性Wistar大鼠分为3组:正常对照组(N组),TAA组,TAA+DSS组。检测实验各组血浆内毒素、丙氨酸转氨酶(ALT)、乳酸脱氢酶(LDH)水平的变化判定肝功能情况;检测肝组织匀浆血栓素B2(TXB2)、6-酮-前列环素1α(6-keto-PGF1α)的含量判定肝脏微循环变化;HE染色显示肝组织形态学改变,Weigert染色显示肝脏血管内微血栓。结果TAA+DSS组与TAA组相比,ALT、LDH、内毒素水平及TXB2含量均明显减少,6-keto-PGF1α含量明显升高(P<0.05或P<0.01);HE染色显示TAA组可见肝细胞点片状坏死、大量的炎性细胞浸润;TAA+DSS组上述改变均明显减轻;Weigert染色显示TAA组微血栓计数明显增加,TAA+DSS组较前明显减少(P<0.01)。结论丹参素通过降低肠源性内毒素水平,改善肝脏微循环障碍,从而减轻急性重型肝损伤。Objective To study the protective role of danshensu in liver injury and the pathogenesls. Methods Acute liver injury was induced by intragastric thionletamide (TAA). The rats were randomly divided into 3 groups: normal control group (N), TAA treated group (TAA), and TAA plus DSS treated group (TAA + DSS). Liver function was evaluated by ALT, LDH and endotoxin in plasma. The liver homogenate levels of thromboxanes B2 (TXB2)and 6-keto- PGF1α would demonstrate the changes of hepatic microcireulation. HE showed histomorphhology and Weigert's staining showed microthrombi. Results The levels of ALT, I.DH, endotoxin, TXB2 in TAA + DSS group significantly decreased ( vs TAA group, P 〈 0.05 or P 〈 0.01 ), nevertheless the levels of 6 kcto-PGF1α significantly increased ( P 〈 0.05). HE, showed: infiltration of inflammatory cells in TAA + DSS group reduced and damage of hepatocytes significantly lighten ( vs TAA group, P 〈 0.05) ; hepatocytes inflated and sinusoidal narrowed, alarge number of inflammatory cells infiltrated and point or slice nccrosis in TAA group. Weigert's staining showed microthrombi in TAA + DSS group significantly reduced ( vs TAA group, P〈 0.01 ). Conclusion Danshensu protects the liver from injury by the decrease of endotoxin amount and the improvement of hepatic microcirculation.
