摘 要:研究猪囊虫DNA疫苗pVAX-S-Δc-3n在仔猪体内的组织分布情况,并对其安全性进行分析。将猪囊虫DNA疫苗以肌注方式免疫仔猪,分别于接种后1 d,7 d,4 w,8 w,12 w分离各组织,提取组织总DNA,用PCR法扩增质粒上抗原基因的部分序列,分析质粒DNA在不同组织内的分布及随时间变化的情况。结果表明接种后第一天,除了脑,其余组织都能检测到质粒,但随时间延长,仅注射部位能检测到质粒,且数量不断下降。肌注质粒在猪体内注射部位以外的组织中较短时间就被清除,因此质粒与染色体发生整合的可能性不大。This paper is to explore the tissue distribution and safety in porcine immunized with a DNA vaccine named pVAX-S-△c-3n. The plasmid vaccine was intramuscularly injected into a new born porcine. After 1 d,7d,4w,8w, 12w, various tissues were removed from porcine, respectively. Total DNA was extracted. PCR was used to amplify the special sequence of concerned antigen gene included in the plasmid and to study the distribution and kinetics of the vaccine in various tissues. Results showed that plasmid was detected in all the tissues except brain 1 d after immunization. But with time going, plasmid was only found in injected sites, with quantity declining. Intramuscularly injected plasmid was cleared in short time except for sites of administration. There is little possibility for plasmid vaccine to integrate into the host genome.
