摘  要:目的研究阿糖胞苷聚氰基丙烯酸正丁酯纳米粒(ARA-C-PBCA-NP)冻干针剂在家兔体内的药物动力学规律。方法以阿糖胞苷(ARA-C)溶液剂为参比,采用HPLC法测定ARA-C-PBCA-NP冻干针剂和ARA-C溶液剂在家兔体内的血药浓度,计算药动学参数。结果ARA-C-PBCA-NP冻干针剂和ARA-C溶液剂在家兔体内的药物动力学过程均符合二室模型,ARA-C-PBCA-NP冻干针剂在家兔体内的T1/2Β和平均滞留时间(MRT)延长,清除率(CL)降低(P<0.05)。结论ARA-C-PBCA-NP冻干针剂可显著延长ARA-C在体内的存留时间,具有明显的缓释特征。Objective The pharmacokinetics of Cytarabine Polybutylcyanoacrylate Nanoparticles (Ara-C-PBCA-NP) lyophilization injection in rabbits was studied. Methods Ara-C water solution was taken as reference, the concentration of Ara-C was determined by HPLC, and the pharmaeokinetic parameters were calculated. Results Pharmaeokinetie properties of Ara-C water solution and Ara-C-PBCA-NP lyophilization injection complied with two-department model, The parameters t1/2β and MRT of Ara-C-PBCA-NP lyophilization injection were prolonged, and CL was reduced dramatieally(P〈0. 05). Conclusion Ara-C-PBCA-NP lyophilization injection possesses prolonged retention time in vivo and significant sustained releasing character.
