摘  要:目的:探讨双歧杆菌DNA对小鼠骨髓树突状细胞(BMDC)成熟的影响.方法:纯化提取双歧杆菌DNA,并鉴定DNA制品CpG基序甲基化程度;FACS检测BMDC的吞噬功能和表面标志分子,ELISA法检测BMDC分泌细胞因子水平,3H-TdR掺入法检测T淋巴细胞增殖能力.结果:双歧杆菌DNA诱导24 h后BMDC高表达Ia,CD40,CD86和CD11 c分子,明显增加IL-6,IL-12(p40),TNF-α的分泌,显著增强同种异体和自体T细胞增殖的能力.结论:双歧杆菌DNA能够促进BMDC成熟,增强BMDC的抗原提呈能力.AIM: To explore the effect of bifidobacteria DNA containing unmethylated CpG motifs (CpG DNA) on the maturation of bone marrow-derived dendritic cells ( BMDC ). METHODS: The genomic DNA of bifidobacteria was extracted and purified, and the methylated degree of CpG motifs was tested. Cell phenotypes and antigens uptake of BMDC were analyzed by flow cytometry. The levels of IL-6, IL-12(p40) and TNF-α were detected by ELISA and T cell proliferation was detected by ^3 HTdR incorportation test. RESULTS: Treated with bifidobacteria DNA for 24 h, BMDC expressed a high level of Ia, CD40, CD86 and CD11 c molecules, significantly increased the secretion of IL- 6, IL-12 (p40) and TNF-α and significantly enhanced the proliferation of allogenic and homogenic T cells. CONCLUSION: Bifidobacteria DNA has remarkable stimulatory effects on BMDC maturation and antigen-presenting function.
