摘 要:目的:在结扎大鼠冠脉建立心肌梗死模型的基础上,通过4,6-联脒-2-苯基吲哚标记骨髓单个核细胞,初步观察外周静脉植入与局部注射植入骨髓单个核细胞对心脏功能恢复的疗效差异,及其在心肌梗死区域的分布情况。方法:实验于2002-01/2003-10在中南大学湘雅医学院生理学系血液生理研究室完成。选取清洁级近交系3月龄Wistar大鼠40只,随机分为正常对照组、心肌梗死对照组、静脉注射移植组、局部注射移植组,10只/。除正常对照组外,其余组建立冠脉结扎心肌梗死模型。术后1组周,心肌梗死对照组不给药,静脉注射移植组静脉注射骨髓单个核细胞,局部注射移植组局部注射骨髓单个核细胞。术后1个月通过心功能检测比较不同移植方式对整体心功能恢复的差异,镜下观察移植的4,6-联脒-2-苯基吲哚标记细胞在心肌梗死区的分布特征及病理形态学变化,采用图像分析系统评价各组左心室心肌梗死面积、心室壁厚度、新生血管的变化。结果:实验选用大鼠40只,全部进入结果分析。①骨髓单个核细胞静脉移植在心肌梗死大鼠体内的分布:大部分布在心肌梗死区及其周围,脾脏以及骨髓也有分布,而肺、肝、肾等组织器官中未发现标记细胞。②术后1个月心肌梗死区移植细胞的鉴定:静脉注射的骨髓单个核细胞细胞核呈蓝色,细胞数目少;局部注射的骨髓单个核细胞细胞核呈蓝色,细胞数目多。③术后1个月各组大鼠心肌组织切片的病理形态学观察:正常对照组心肌排列有序,未见坏死等改变;心肌梗死对照组心肌组织明显纤维化,心室壁明显变薄;静脉注射移植组心室壁变厚,恢复的心肌位于梗死区中心和/周边;局部注射移植组新生和或恢复的心肌位或于梗死区中心和/或周边,心室壁明显变厚。④术后1个月不同移植方式对心肌梗死大鼠血流动力学的影响:与正常对照组比较,心肌�AIM: To primarily observe the different efficacy of peripheral venous transplantation and local injection of rat bone marrow mononuclear cells (rBMNCs) on the recovery of cardiac function, and the distribution in the myocardial infarcted areas on the basis of establishment of myocardial infarction models by ligation of coronary through labeling rBMNs by 4,6-diamidino-2-phenylindole dihydrochloride (DAPI). METHODS: The experiment was carried out in the Research Laboratory of Blood Physiology, Department of Physiology, Xiangya Medical College of Central South University between January 2002 and October 2003. Forty Wistar rats of 3 months old were randomly divided into 4 groups with 10 rats in each group: normal control group, myocardial infarction control group, vein transplantation group and endomyoeardial injection group. Except the normal control group, the rats in the other groups were made into models of myocardial infarction by left anterior descending occlusion. One week after operation, no administration was given in the myocardial infarction control group, and the rats in the vein transplantation group and endomyocardial injection group were treated with intravenous injection and local injection of rBMNCs respectively. One month after operation, the differences of recovery of general cardiac function by different transplanted methods were compared by the detection of cardiac function, the distributive features of DAPI-labeled various cells in the myoarcardial infarcted area and the pathomorphological changes were observed under microscope. The left ventricular myocardial infarcted area, thickness of ventricular wall and angiogenesis changes in the groups were evaluated with the image analysis system. RESULTS: All the 40 rats were involved in the analysis of results.① Distribution of DAPI-labeled cells in the rat's body: the DAPI-labeled cells with DAPI were mainly found at the infarction zone, at the bone marrow and spleen also, and not at lung, liver and kidney tissue. ②Evaluation
