摘 要:目的:介绍猪膀胱无细胞基质的制备方法并观察其与异种血清是否存在抗原抗体结合反应。方法:实验于2002-01/2003-01在解放军第三军医大学大坪医院野战外科研究所全军“三优”实验室完成。新鲜贵州香猪3只,其膀胱用生化方法脱细胞制备膀胱无细胞基质;分别以人AB,O,A,B型、大鼠、兔血清为一抗,免疫组织化学方法观察猪正常膀胱组织、膀胱无细胞基质分别对不同血清的结合情况。结果:实验贵州香猪3只均进入结果分析。①用生化脱细胞方法可成功制备富含胶原等细胞外基质的膀胱无细胞基质。②由贵州香猪全层膀胱制备的膀胱无细胞基质对异种血清的抗原抗体反应均为阴性;正常膀胱组织与异种血清均存在抗原抗体结合反应,主要发生于膀胱固有层及肌间隙的血管壁。结论:贵州香猪膀胱可用生化脱细胞方法制备以细胞外基质成分为主的组织工程天然材料;制备成功的膀胱无细胞基质已不存在可能导致异种移植后超急性排斥反应的靶抗原。AIM: To introduce the making procedure of porcine bladder acellular matrix (PBAM) and explore whether the binding reaction of antigen and antibody with xenogeneic serum existing or not. METHODS: The experiment was conducted at the "three good" laboratory, Institute of Battle Surgery, Daping Hospital, Third Military Medical University of Chinese PLA between January 2002 and January 2003. PBAM were extracted from the fresh bladders of 3 Guizhou small pigs using the biochemical procedures by the removal of cellular components. The sera of human (blood type AB, O, A and B) were used; rats and rabbits as primary anti-bodies, the binding status of normal bladder tissue and PBAM with different serum was observed with immunohistochemical method. RESULTS: The experimental 3 Guizhou small pigs were all involved in the result analysis. ①PBAM that contained rich collagen could be gained with biochemical procedures by the removal of cellular components. ②Negative stained results were obtained from the PBAM made by whole bladder of Guizhou pig against xenogeneic sera. Binding reaction of antigen and antibody existed between normal bladder tissue and xenogeneic serum, which occurred often at lamina propria of urinary bladder and dissepiment of muscle interspace. CONCLUSION: Bladders of Guizhou pigs can be made into prospective natural tissue engineering materials that mainly composed of extracellular matrix with biochemical procedures by the removal of cellular components. In establishing the PBAM successfully, there are not target antigens that may cause hyperacute rejection 'after xenotransplantation.
