摘 要:目的:转录因子激活蛋白1中的重要亚单位c-fos与软骨细胞增殖、分化以及细胞的生理进程和炎症等疾病密切相关。资料来源:应用计算机检索PUBMED2000-01/2005-01和EBSCO2000-01/2005-01期间的相关文章,检索词为“c-fos”,并限定文章语言种类为英文。同时计算机检索万方数据库2000-01/2005-01期间的相关文章,检索词为“原癌基因,立早基因”,并限定文章语言种类为中文。资料选择:对资料进行初审,以关键词“软骨,增殖,分化,炎症”为条件对所获文献进行二次检索。纳入标准:文章应涉及c-fos基因与软骨细胞增殖、分化及炎症的内容。排除标准:重复研究或Meta分析类文章。资料提炼:共收集到24篇相关文献,17篇文献符合纳入标准,排除的7篇文献是由于内容陈旧或重复。资料综合:对剩余的17篇文献全文中的相关信息进行分类综合研究。软骨细胞发育成熟过程中,在各种生长因子和外界环境变化作用下,会发生明显形态和生化成分的改变。在这一过程中,作为细胞信号通路上的信使之一的c-fos起到了重要的作用。过度的表达c-fos会诱发软骨肉瘤,软骨细胞不能正常成熟,而去除c-fos则肢体发育短小,生长板断裂,生长期细胞减少,肥大带扩大。软骨关节炎早期,软骨表现出一种自我修复的能力,细胞体积变肥大,蛋白聚糖增加。当后期平衡被打破,炎症所致破坏效应则显现出来,其中c-fos基因充当了重要信使。结论:c-fos基因可以结合在许多基因的启动子上参与转录调控,并在转录前后对蛋白结合DNA等多个水平的表达进行调节,与软骨的分化和炎症密切相关,提示可以针对软骨疾病患者开展以c-fos为靶点的药物及基因治疗。OBJECTIVE: The c-fos that is the important sub-unit of the transcription factor activating protein-1 has close relation with proliferation and differentiation of chondrocytes, physiological processes of the cells and inflammation, etc. DATA SOURCES: Using the key words "c-fos", we searched the PUBMED and EBSCO databases for related articles published from January 2000 to January 2005 by computer in English. Meanwhile, we searched the Wanfang database for related articles published between January 2000 and January 2005 with the key words "protooncogene, immediate early gene"in Chinese. STUDY SELECTION: After the first selection, the second retrieve was conducted with the key words "cartilage, proliferation, differentiation, inflammation". Inclusion criteria were articles on c-fos gene and the proliferation and differentiation in ehondrocytes as well as inflammation. Exclusion criteria were articles with repetitive research or Meta analysis. DATA EXTRACTION: Twenty-four related articles were collected. Seventeen were in accordance with the inclusion criteria. The excluded 7 articles were due to old or repeated content. DATA SYNTHESIS: The related contents in the rest 17 literatures were classified and synthesized. During the course of development and maturation, the ehondrocyte displayed great morphological and biochemical change under the control of kinds of growth factors and changeable environments. The c-fos, which was one of important messengers on the signal pathway, played the important role in this process, Over-expression would cause the chondrosareoma and induce abnormal maturation. Loss of function in c-fos knockout would demonstrate a diminished zone of proliferation chondrocytes, disruption of growth plate, acrodysplasia and extension of terminal hypertrophy zone. Osteoarthritis was characterized by progressive deterioration and erosion of joint cartilage.
