摘 要:目的确定更多的与肝再生相关的基因,方法借助4~8正向抑制性消减文库(0-4-8-12短间隔连续部分肝切除)产生的α2-巨球蛋白基因片段.运用cDNA芯片分析了其mRNA在再生肝中的表达动态。结果肝再生中α2-巨球蛋白呈上调表达.而且变化幅度很大,至高点超过21倍。大部分肝切除中α2-巨球蛋白的表达高峰出现在8h和16h.短间隔连续部分肝切除中α2-巨球蛋白的表达呈现不同的变化趋势.4h短间隔的连续肝切除诱导α2-巨球蛋白表达不断升高.而第二次的36h短间隔的连续肝切除才能诱导α2-巨球蛋白表达显著升高。结论肝再生中α2-巨球蛋白可能在早期的正相急性反应中起重要作用,减少切除伤害带来的炎症.促进肝再生的顺利进行。Objective To identify more genes related to liver regeneration bv using eDNA miero assay ( rat liver eDNA chip). Methods This study analyzed expression change of alpha-2-maeroglobulin mRNA from 4-8 forward (0-4-8-12 h short interval successive partial hepatectomy) subtractive eDNA libraries in regenerating rat liver. Results Alpha-2-maeroglobulin was largely up-regulated in liver regeneration and was more than 21 times normal level at the expression peak. Expression peaks of alpha-2-macroglobulin appeared 8 h and 16 h after partial hepatectomy (PH). Expression change of alpha-2-maeroglobulin during SISPH presentsed different profiles in contrast to that of PH. Successive hepatectomy with 4 h interval could induce successive increase of alpha-2-macroglobulin and only the second successive hepatectomy after 36 h could induce marked increase. Conclusion The alpha-2-maeroglobulin might play an important role in early stress response in alleviating inflammation induced by hepatectomy and ensuring liver regeneration.
