摘  要:为揭示SARS相关冠状病毒(SARS—associated coronavirus,SARS-CoV)的复制特点,并探讨该病毒的致病机制,利用负染电子显微镜(EM),超薄切片EM和免疫EM技术研究了SARS-CoV在Vero细胞E的形态学特征。结果显示,成熟病毒颗粒多为圆形或椭圆形,直径80~120nm,包膜上有放射状排列的纤毛样突起,长约20nm,基底窄。此外可见哑铃形、肾形及“钉子样”等多形性成熟病毒颗粒,这屿颗粒能与病人恢复期血清和抗S蛋白抗体反应。细胞内病毒颗粒多位于包涵体内,呈显著的多形性,直径为20~400nm,其形状可分为:①圆形、肾形或椭圆形;②管样结构;③不规则形,如三角形、哑铃形等,另外可见一种特殊的分枝样颗粒。颗粒电子密度不一,有实心和空心两种,其中守心颗粒周边的电子密度高。还观察到螺旋形的病毒核衣壳结构。In order to explore the rnorphogenesis linked to the replicating mechanism of SARS-associated coronavirus (SARS-CoV), a comprehensive electron microscopy (EM), including negative staining, ultra-thin sectioning and immune colloidal gold EM, was made in fully use. A striking polymorphism of the SARS-CoV both in shape and size in Vero cell cultures attracted the authors attention. The size variation was remarkable ranking from 400nm to 20nm, with size decreasing tendency in immature or provirions parallel to the increasing of density in mature virions. The mature virions were more or less spherical in appearance. They were morphologically identical to the ordinary coronaviruses, whereas, the variation of the immature virions was more eminent. Along with the ordinary ones, there were some aberrant particles dispersing within the cytoplasm of the infected cells such as kidney-like, dumb bell-like, and nail-like particles. Occasionally, branch-like virions were also distinctly seen. What was more, some unusual figure of the nucleoprotein-like structures could be visualized in the cytoplasmic matrix of the infected cells. This structure looked like zigzag, most probably representing the helix of the genomic component of the virus.
