摘 要:目的:了解骨延长过程中BMP2基因在骨延长区组织细胞的表达和定位,从分子水平探讨延长区骨修复的机制。方法:采用兔胫骨上干骺端截骨延长动物模型,24只新西兰大白兔分8个时相点取材。通过延长区组织石蜡切片,BMP2地高辛标记的mRNA探针原位杂交。结果:BMP2mRNA阳性表达持续到延长结束后20d,表达主要定位于延长区假性生长板中的成纤维细胞样间充质细胞、成纤维细胞、骨膜生发层的骨形成细胞及骨小梁边缘的成骨细胞,并随着骨延长不同阶段而有所变化。达到预定延长长度20d以后,延长区BMP2mRNA表达逐渐转为阴性。结论:骨延长区BMP2mRNA的表达和细胞定位与骨折愈合不同。BMP2在延长区高水平、长时效的表达对其骨修复具有重要作用,使延长区骨组织能够持续形成,短时间内以骨再生形式修复大段延长间隙。Objoctive: To study and discuss the mechanism of the bone regeneration during limb lengthening at the molecular level. Methods: Upper tibia metaphyseal lengthening model was prepared in 24 New Zealand rabbits, All animals were randomly divided into 8 groups and the distracted bones were got at the scheduled time. Slices were made and the expression and cellular localizations of BMP2 mRNA were investigated with in situ hybridization (ISH). Results: Positive expression of BMP2 mRNA was detected to last to 20 days after lengthening, and localized mainly in the pseudo-plate area, especially in fibroblast-like mesenchymal cells, fibroblasts, osteoblasts under periosteum and osteoblasts around bone trabecula, and gradually disappeared after 20 days of lengthening. Conclusions: The expression and localization of BMP2 gene in the experimental bone lengthening area is different from that in fracture healing. The high level and long time expression in the distracted area plays an important role on bone repairing in the lengthening, it continually maintains bone forming ability, so in relative short time the bone regeneration could refill the large segment of bone gap in distracted area.
