摘 要:目的评价bcl-2反义硫代磷酸寡核酸(ASODN)对骨髓造血功能的影响。方法正常骨髓与含终浓度为10 mol/L的bcl-2 ASODN共培养12 h,以处理后的骨髓细胞为反义组;空白对照组除不加ASODN外处理与反义组相同。分别以2组骨髓细胞建立短期与长期培养体系,动态观察粒、单核系集落形成单位(CFU-GM)的形态和产率;培养结束时,非贴壁细胞以流式细胞仪检测CD34和bcl-2蛋白表达,并观察造血细胞和造血基质的生长态势。结果CFU-GM的形态和产率、CD34+细胞的bcl-2蛋白表达反义组均与空白对照组相近,造血细胞和造血基质的生长态势良好。结论在正常骨髓体外培养条件下,bcl-2 ASODN不影响骨髓细胞的造血功能,可能用于白血病骨髓的体外净化。Objective To demonstrate that antisense bcl-2 oligodeoxynucleotides was nontoxic to hematopoietic progenitor cells. Methods Normal bone marrow cells were incubated with and without 10 μmol/L antisense bcl-2 oligodeoxynucleotides for 12 hours. The treated cells were collected as antisense group and blank control group and used for short-and long-term culture. The hematopoietic progenitor cell survival were evaluated by the growth of myeloid (CFU-GM) progenitor cells in both short-term and long-term cell cul- ture the percentage of the coexpression of bcl-2 and CD34^+ molecules were examined by dual-color flow cytomettic analysis. The long-term culture-initiating cell activity were observed under microscope. Results Comparing to the control, no significant effect of antisense bcl-2 oligodeoxynucleotides on CFU-GM growth and any change of the coexpression of bcl-2 and CD34^+ was observed. Under the treatment of antisense bcl-2 oligodeoxynucleotides, the hematopoietic cells and the corresponding fibroblasts grew morphologically well in the long-term culture system. Conclusion Antisense bcl-2 oligodeoxynucleotides is nontoxic to hematopoietic pro- genitor cell under in vitro culture condition, which suggested it can be safe for bone marrow purifying.
