细胞色素芳香化酶基因及环氧合酶-2基因mRNA在子宫内膜异位症异位与在位内膜的表达

摘　　要：目的探讨细胞色素芳香化酶(P450arom)基因、环氧合酶-2(COX-2)基因mRNA在子宫内膜异位症(内异症)患者异位、在位子宫内膜组织中的表达及其意义。方法采用逆转录聚合酶链反应(RT-PCR)检测内异症患者在位及异位子宫内膜及正常子宫内膜中P450arom mRNA、COX-2 mRNA的表达情况。结果P450arom mRNA在内异症患者异位及在位子宫内膜均有强表达,正常子宫内膜无表达,分泌期、增殖期无差别;COX-2 mRNA在内异症患者异位及在位子宫内膜均有强表达,正常子宫内膜弱表达(P<0.05),且内异症患者在位子宫内膜及正常子宫内膜COX-2 mRNA表达分泌期均略强于增殖期,但差别均无统计学意义(P>0.05),异位内膜分泌期、增殖期无差别。不同rAFS分期内异症子宫内膜P450arom mRNA、COX-2 mRNA的表达无差别。结论子宫内膜P450arom mRNA、COX-2 mRNA的高表达与内异症的发生、发展可能密切相关。Objective To study the expression and significance of aromatase cyclooxygenase-2 P450 mRNA and cytochrome in the ectopic and eutopic endometrium of women with endometriosis. Metheods Reverse transeript-polymerase chain reaction（RT-PCR） was used to examine P450 arom mRNA and COX-2 mRNA expression. Results Expression of P450 arom mRNA and COX-2 mRNA in the ectopic and eutopic endometrium of women with endometriosis were strongly positive, whereas the level of COX-2 mRNA was low and no expression of P450 arom mRNA was found in the normal endometrium. Expression of P450arom mRNA in the ectopic endometrium was no variation in regard to menstrual phases. The level of the expression of COX-2 mRNA in the secretory phase was higher than that in the proliferative phase in both endometrium from women with and without endometriosis but are of no significance between two groups, however, it remained significantly high in the ectopic endometrium of women with endometriosis regardless of the menstrual phase; There was no difference between the expression of both mRNA and rAFS staging of endometriosis. Conclusions High level of expression of P450 arom mRNA and COX-2 mRNA may be closely related in the ectopic and eutopic endometriasis of women.
