摘 要:目的研究葛根芩连加味汤对糖尿病大鼠视网膜病变的防治作用及可能的药理作用。方法实验大鼠随机分为正常对照组、用药糖尿病组和不用药糖尿病3组,分别记录血糖,16周测糖化血红蛋白,观察光镜和电镜下视网膜、视网膜铺片的血管形态及细胞计数。结果两组糖尿病大鼠均在注射佐链尿菌素(STZ)1周后血糖升高。出现了糖尿病的典型症状及白内障,但用药组症状相对较轻。糖尿病大鼠的糖化血红蛋白值比正常组升高,不用药组比用药组升高更显著,差异有统计学意义(P<0·01)。糖尿病组比正常组视网膜血管内皮细胞/周细胞(E/P)值有所上升,不用药组表现较用药组严重,差异有显著性(P<0·01)。电镜观察发现糖尿病不用药组大鼠视网膜血管基底膜增厚、血管闭塞。结论葛根芩连加味汤有改善糖尿病大鼠视网膜早期病变的作用,可能与抑制非酶糖基化终末产物及抑制醛糖还原酶相关。OBJECTIVE To investigate the prevention and cure action of the traditional Chinese medicine gegenqinlianjiaweitang (葛根芩连加味汤) to diabetic rat retinopathy.METHODS Thirty adult male rats were divided into three groups, 10 rqts each. Diabetes were induced in rats of groups 2 and 3, respectively by abdominal cavity injection of streptozocia (60mg/kg) .The group 2 rats were treated with the traditional Chinese medicine gegenqinlianjiaweitang every day. Rats were observed for 16 weeks including blood glucose and eye manifestation. At the end of 16 week ferrohemoglobin were measureed. Then killed rats. Six randomly selected rats in each group were enucleated one eye for retinal preparation. The retinal vessels and numbers of pericytes and endothelial cells were evaluated via light microscope and electron microscope in two eyes of each groups. RESULTS All diabetic rats demonstrated hyperglycemia and hih ferrohemoglobin. But the ferrohemoglobin of untreated rats are higher than the treated group (P 〈 0.01 ) .The endothelial/pericyte ratio in diabetic rat was higher than that in nondiabetic rats. The alter of untreated diabetic rat were significant higher than that in treated diabetic rats ( P 〈 0.01 ) . In addition, the untreated diabetic rat group illustrated thickening retina blood vessel basement membrane and segment of blood vessel occluded. CONCLUSIONS The gegenqinlianjiaweitang can ameliorate early retinopathy of diabetic rat. The action of the medicine maybe relate to antiadvanced glycation end products and anti - aldose reductase.
