摘  要:以甘蓝型油菜品种N1、N2、N3、N4、N5和N6的下胚轴为材料,研究外植体芽再生的影响因素.结果表明:不同植物激素的组合、Ag+、蔗糖浓度、GA3、外植体的生理状态即苗龄以及基因型对外植体的再生均有较大影响.4 d苗龄的下胚轴置于含1.5 mg?L-12,4-D的MS培养基中预培养7 d,再转接到含30 g?L-1蔗糖的分化培养基(MS+BA 4 mg?L-1+NAA 0.1 mg?L-1+AgNO32.5 mg?L-1)上,芽再生频率和再生外植体数均较高,其中品种N1分别达到了40%和3.7.添加1.0 mg?L-1GA3可促进胚状体形成,芽苗再生频率显著提高(品种N1达到了95%,比不添加GA3提高了1倍多),但较易形成玻璃苗.Effects of various hormones, Ag^+, sucrose concentration, GA3, seedling age and genotypes on shoot regeneration from hypocotyl segments of 6 varieties of Brassica napus were examined in vitro. Results showed that all these factors had great influence. Hypocotyl explants of 4-day-old seedlings showed high regeneration frequency and number of shoots per explant (40% and 3.7 in cv. N1 ), which were pre-cultured for 7 days in a MS medium with 1.5 mg ·L^-1 2, 4-D and then sub-cultured in differentiation medium of MS+4 mg·L^-1 BA+0.1 mg·L^-1 NAA+2.5 mg·L^-1 AgNO3 supplemented with 30 g·L^-1 sucrose. Addition of 1.0 mg·L^-1 GA3 in media could promote the formation of embryos and improve shoot regeneration frequency remarkably (reached 95% in cv. N1 ). Vitrification of the plantlets was, however, more liable to occur under such condition.
