Influence of Radix Astragali on nitric oxide and endothelin-1 in pulmonary tissue in hypoxemic pulmonary hypertension in rats

摘  要:目的探讨黄芪注射液对低氧性肺动脉高压(HPH)大鼠肺组织中内皮素1(ET-1)和一氧化氮(No)的影响。方法将体重为210—310g健康雄性Wistar大鼠21只随机分为3组,每组7只。(1)对照组:室内空气正常饲养;(2)低氧组:在常压低氧舱内[氧浓度(10.0±0.5)%]缺氧,每天8h,每周6d,连续30d;(3)治疗组:缺氧条件同低氧组,自缺氧第一天起给大鼠每天腹腔内注射8g/kg黄芪注射液。对照组与低氧组每天腹腔内注射生理盐水量和治疗组相同。于低氧30d后用右心导管法测定肺动脉平均压(mPAP),测定颈动脉平均压(mCAP),分光光度法测定肺组织中NO含量和放射免疫法测定肺组织中ET-1含量,观察肺中、小肌型动脉超微结构的变化情况。结果(1)低氧组mPAP(rainHg)(21.9±1.6)、ET-1(ps/ml)(309.1±58.1)明显高于治疗组(16.2±0.8、287.7±57.5)和对照组(15.3±0.8、241.1±52.5)(P〈0.01和〈0.05),而治疗组与对照组相比差异无统计学意义。(2)大鼠肺组织中NO含量(μmol/L)低氧组(6.5±0.3)明显低于治疗组(7.9±1.2)和对照组(9.2±0.9),差异有统计学意义(P〈0.01),治疗组虽高于低氧组,但仍低于对照组(P〈0.05)。(3)三组之间mCAP比较差异无统计学意义。(4)电镜下可见:对照组肺小动脉内皮细胞扁平、结构正常,低氧组肺中、小肌型动脉内皮细胞增生、肥厚、肿胀,平滑肌细胞肥厚,胞浆内可见大量线粒体和内质网,治疗组内皮细胞趋于扁平、胞浆内空泡明显减少。结论慢性低氧可引起大鼠肺组织中ET-1含量升高、NO含量下降、肺血管结构重建及内皮损伤,导致mPAP上升,黄芪注射液可调节肺组织中ET-1、NO含量,对HPH有良好的治疗作用。Objective To study the influence of Radix Astragali (RA) on pulmonary tissue endothelin-1 (ET-1) and nitric oxide (NO) in hypoxic pulmonary hypertension rats. Methods Twenty one healthy male Wistar rats weighing 210-310 g were divided into three group at random with 7 in each. The rats in control group were raised in ordinary room condition; those in hypoxic group were raised in ordinary pressure hypoxic box [concentration of 02 was( 10. 0 ±0. 5)% ] for 8 hours a day, for 30 days; those in RA group were raised in the same condition as hypoxic group and treated with an intraperitoneal injection of RA 8 g/kg per day. The rats in the control group and hypoxic group were given the same volume of intraperitoneal injection of normal saline. Mean pulmonary arterial pressure (mPAP), mean carotid artery pressure (mCAP) were measured via right cardiac catheterization, concentration of NO in pulmonary tissue was measured by radioimmunoassay. Results (1) The mPAP (mmHg)( 21.9±1.6 ) and ET-l(pg/ml) (309. 1 ±58. 1) in hypoxemic group were significantly higher than those in RA group ( 16. 2 ±0. 8,287. 7 ± 57.5) and control group ( 15. 3 ± 0. 8,241.1 ± 52.5 ) ( P 〈 0. 01, 〈 0. 05 ), but the difference between RA group and control group was not significant. (2) NO(μmol/L) in pulmonary tissue in hypoxemic group (6. 5 ± 0. 3 ) was lower than that in RA group and control group (9.2 ± 0. 9), NO in RA group was higher than that in hypoxic group but lower than that in control group (P 〈 0. 05). (3) There was no significant difference in reCAP among the three groups (P 〉 0. 05). (4) Under electron microscope, the endothelial cells of arterioles of the lung tissue of control group were flat and had normal morphology. However, in the lung tissue of hypoxie group, there were proliferation, hypertrophy and swelling of endothelial cells of pulmonary medium and small arteries and plenty of mitochondria and endoplasmic reticula in cytoplas
