摘 要:目的探讨嗅鞘细胞对脊髓源性神经干细胞分化的作用。方法应用M illicell插入式细胞培养皿,大鼠脊髓源性神经干细胞与嗅鞘细胞在无血清培养液中共培养,通过免疫化学染色方法,检测脊髓源性神经干细胞分化为神经元、星形胶质细胞和少突胶质细胞的比例。结果共培养7 d后,脊髓源性神经干细胞分化为神经元的比例高达43.5%,明显高于对照组的21.4%。结论嗅鞘细胞提供的微环境可促进脊髓源性神经干细胞分化为神经元。Objective To investigate the role of olfactory ensheathing cells in differentiating spinal cord-derived neural stem cells (NSCs). Methods By using Millicell cell culture plate insert, rat spinal cord-derived NSCs were co-cultured with olfactory ensheathing cells in the serum-free medium. The proportion of neurons, astrocytes and oligodendrocytes differentiated by spinal cord-derived NSCs was investigated by means of immunochemical staining. Results At day 7 of co-culture, the proportion of spinal cord-derived NSCs differentiating into neurons was as high as 43.5%, which was significantly higher than 21.4% in control group. Conclusions The environmental cues provided by olfactory ensheathing cells can promote the differentiation of co-cultured spinal cord-derived NSCs into neurons.
