摘  要:目的观察银杏叶提取物(EGb761)对大鼠视网膜神经细胞线粒体膜电位(MMP)的影响及其对抗谷氨酸兴奋性毒性对视网膜神经细胞的保护作用。方法培养视网膜神经细胞,用Rhodamine123标记培养7d的视网膜神经细胞线粒体,分为正常对照组、EGb761组、谷氨酸组以及EGb761+谷氨酸组,共聚焦激光显微镜动态检测MMP的变化。结果与正常对照组比较谷氨酸组MMP下降;EGb761组MMP及EGb761+谷氨酸组MMP升高,P<0.01。结论EGb761可能通过升高视网膜神经细胞MMP的途径影响线粒体的功能;并且可对抗谷氨酸兴奋性毒性,保护视网膜神经细胞。Objective To investigate the effect of EGb761 on mitochondria membrane potential and glutamate-induced excitotoxicity in primary cultured rat retinal neurons. Methods Retinas of twenty 2-6-day old Sprague-Dawley rats were isolated and dissociated by enzyme and mechanical method. The cells were cultured in 95% O2-5% CO2 incubator for 7 days. Then the retinal neurons were treated with culture media,glutamate, EGb761 and EGb761 + glutamate separately. Mitochondria membrane potential of the neurons was determined on day 7 by laser scan confocal microscopy using Rhodamine123 as indicator. Results The mitochondria membrane potential of the neurons in the control group was 9.81±5.60 in 1 minute,9. 12±5.54 in 3 minutes and 9.37±5.58 in 5 minutes. Compared with control, the mitochondria membrane potential of the neurons was markedly reduced from 1 minute to 5 minutes (5.82±3.32 in 1 minute, 5.75± 3.72 in 3 minutes and 6. 11±4.02 in 5 minutes)from exposure to lmmol/L glutamate,while that of EGb761 group was significantly increased from 1 minute to 5 minutes ( 18.59±9.33 in minute,60. 94±36.80 in 3 minutes and 69.02±43.65 in 5 minutes) in comparison with control group and glutamate group( P 〈0.01 ,P 〈 0.01 ) and a similar result was obtained in the EGb761+ glutamate group ( 16.61 ±9.97 in 1 minute,15.55±10.82 in 3 minutes and 17.92±7.62 in 5 minutes) (P〈0.01,P〈0.01). Conclusion The results suggest the effect of EGb761 on retinal neurons might be due to its ability to increase mitochondria membrane potential and EGb761 can protect retinal neurons against glutamate induced neurotoxicity.
