摘 要:目的:探讨叶酸缺乏孕鼠的子代心脏发育过程中GATA-4基因和蛋白的表达改变。方法:成熟雌性SD大鼠36只随机分为实验组和对照组各18只,分别喂以缺乏叶酸和添加叶酸的纯合饲料。两周后与成熟SD雄性大鼠交配,分别取孕13.5天(E13.5d)、孕17.5天(E17.5d)胚胎的心脏及新生鼠的心脏。用RT-PCR检测GATA-4基因mRNA的表达。Western-blotting测GATA-4蛋白水平的表达。结果:GATA-4基因mRNA在E13.5d、E17.5d的胚胎心脏及新生鼠心脏中的表达量,实验组均显著低于对照组(P<0.05)。GATA-4蛋白在E13.5d、E17.5d的胚胎心脏及新生鼠心脏中的表达量,实验组均显著低于对照组(P<0.05)。结论:叶酸的缺乏影响GATA-4基因和蛋白水平的表达,可能导致心脏发生发育中形态的改变,从而造成心脏的功能缺陷。Objective: To study the expression of GATA-4 in the heart of offspring during the development of embryo, when the mother is deficient of folic acid. Methods: Thirty-six adult female so rats were chosen and allorated randomly into control group(18) and study group (18). The control group received non-folic acid fodder and the study group received folic acid fodder for two weeks. Then the female rats in two groups copulated with normal male rats. The hearts were collected from the 13.5-day, 17.5-day embryos and the newborns. GATA-4 mRNA expression was measared by RTPCR and GATA-4 protein was measured by Western-blotting. Results: The expression level of GATA-4 mRNA and GATA-protein in the study group was lower compared with the control group (P 〈 0.05). Conclusion: The changes of the expression of GATA-4 mRNA and protein are due to folic acid deficiency during the pregnancy.
