摘 要:目的观察早期糖尿病大鼠肾脏肌球蛋白轻链激酶(MLCK)表达及意义。方法腹腔注射链尿佐菌素(65mg/kg)诱发糖尿病模型,应用胰岛素控制血糖,心脏取血检测血糖及肾功能相关指标。通过免疫组化与Western-blot检测肾组织肌球蛋白轻链激酶表达。结果与正常对照组相比,模型组血糖及肾功能相关指标发生显著性变化。正常组肾小球、肾小管及肾脏血管均有MLCK的表达。模型组与正常对照组相比,肾小球表达MLCK有一定的减弱,治疗组与正常对照组相比无明显变化。结论早期糖尿病肾病形成可能是由于肌球蛋白轻链激酶表达降低所致。胰岛素治疗糖尿病肾病机制可能是增加肾脏肌球蛋白轻链激酶的表达。Objective To study the expression and significance of myosin light chain kinase(MLCK) in the kidney of diabetic rats. Methods Diabetes was induced by intraperitoneal injection of streptozotocin( STZ 65 mg/kg body wt). Blood glucose were measured by an autoanalyser . The expression of MLCK was analyzed in renal tissue by immunohistochemistry and western-blot methods respectively. Results Plasma glucose of model group was significantly changed compared with that of the control group. The level of the MLCK was lower in renal tissue of 4-week diabetic rats than that in normal rats, whereas the changes were not obvious in kidney of model group treated with insulin in 4-week STZ-induced diabetic rats. Conclusion It is suggested that the formation of diabetic may be closely correlated with the low expression of MLCK in the diabetic rats. Insulin may play an important role in prevention of diabetic by increasing the expression of MLCK.
