摘 要:目的研究胎儿毛乳头细胞的体外成骨能力,为骨组织工程提供一种新的种子细胞。方法Ⅰ型胶原酶消化法分离毛乳头,常规成纤维细胞培养液培养毛乳头细胞。第3代和第7代细胞经成骨诱导液诱导培养7d,以免疫荧光、VonKossa等方法鉴定毛乳头细胞及检测细胞的成骨特性。结果培养的毛乳头细胞呈聚集性生长,表达平滑肌肌动蛋白和Ⅰ型胶原。经成骨诱导后表达骨桥蛋白,有钙结节形成。结论毛乳头细胞具有成骨活性,可作为骨组织工程的种子细胞。Objective To explore the osteogenic potential of cultured fetal dermal papilla cells in vitro and provide a new ideal seeded cells for bone tissue engineering. Methods Dermal papilla were isolated by the digestion method of collagenase I and cultured in routine fibroblast medium. Identified the osteogeneric capability of the third and seventh passage cells by immunofluorescence and Von Kossa after 7 clays of culture in osteogenic medium, Results The cuhiveated dermal papilla cells showed distinct tendency to form clumps at confluence, and expressed high levels of a-smooth muscle actin and type I collagen. Cultured in osteogenic medium, those cells produce calcified deposits and express osteopontin. Conclusion The cultivated dermal papilla cells has osteogeneric capability, and could become a new seeded cells for bone tissue engmeermg.
