摘 要:目的研究妊娠激素对HIV-1体外感染MT-2细胞及复制的影响。方法检测终浓度为1×106U/L的重组人绒毛膜促性腺激素(hCG)和终浓度为10-7mol/L(相当于妇女分泌期外周血水平)、10-6mol/L(相当于妊娠妇女外周血水平)和10-5mol/L(相当于母胎界面局部浓度)的雌二醇(E2)和孕酮(P4)对HIV-1介导荧光标记的HIV-1ⅢB/H9与未标记荧光的MT-2细胞融合的影响;ELISA法检测感染HIV的MT-2细胞培养上清p24抗原含量来探讨妊娠激素对HIV-1复制的影响。结果10-5mol/L的E2和P4可以明显抑制HIV-1介导的细胞融合,合胞体形成数分别为14.87±2.18和16.52±3.20,与对照组(28.93±4.22)比较有极显著性差异(P<0.01),hCG和10-6mol/L的E2也能抑制HIV-1ⅢB/H9和MT-2细胞的融合,融合数分别为22.18±3.87和24.16±4.25,与对照组比较有显著性差异(P<0.05)。hCG及10-5mol/L E2和P4能明显降低感染细胞培养上清中p24抗原含量,依次为(42.61±2.97)pg/ml(、44.28±3.42)pg/ml和(45.37±2.88)pg/ml,与对照组(57.22±3.10)pg/ml比较有显著性差异(P<0.05)。结论妊娠激素hCG、E2和P4体外可以抑制HIV-1感染MT-2细胞及在其内复制。Objective To investigate the effects of pregnant hormones on HIV-1 infecting MT-2 cells and replicating in them in vitro. Methods The effect of Human chorionic gonadotropin (hCG) in final concentration of 1 × 10^6U/L, 17-βestrogen(E2 ) and progesterone(P4) in final concentration of 10^-7mol/L, corresponding levels presented in the circulation during the secretory phase of the cycle, 10^-6mol/L, corresponding peripheral blood levels during pregnancy, and concentration as high as 10^-5mol/L detected at the fetal- maternal interface on HIV-1 infection and replication in vitro, which were detected by HIV-1 mediated cell fusion between MT-2 cells and HIV-1 Ⅲ B/H9 cells labeled calcein-AM and p24 production monitored in culture supematant of MT-2 cells infected by HIV-1 Ⅲ B, respectively. Results E2 and P4 could dramatically inhibit formations of fusion mediated by HIV-1 in concentration of 10^-5 mol/L, and the amount of fusion was 14.87 ± 2.18 and 16.52 ±3.20, respectively. There was a very distinct difference between them and control group, whose amount of fusion was 28.93 ± 4.22( P 〈 0.01). HCG and E of 10^-6mol/L could decrease formation of fusion between HIV-1 Ⅲ B/H9 cells and MT-2 cells too, and the number were 22.18 ± 3.87 and 24.16 ± 4.25 respectively, which were lower than control group (P 〈 0.05). HCG,E2 and P4 of 10^-5 mol/L could decreasing producing of p24, and the concentrations of p24 were (42.61 ± 2.97) pg/ml, (44.28 ± 3.42) pg/ml and (45.37 ± 2.88) pg/ml, respectively, which were lower than that of control group( P 〈 0.05 ). Conclusion HCG, E2 and P4 in given concentration can inhibit the infection and replication of HIV-1 in vitro.
