摘  要:目的探讨人类纤溶酶原激活物抑制物-1(PAI-1)基因4G/5G多态性与2型糖尿病(T2DM)患者股动脉内中膜厚度(FA-IMT)的关系。方法采用等位基因特异性多聚酶链式反应技术检测303例T2DM患者和93例健康个体PAI-1基因4G/5G多态性,利用B型超声检测T2DM患者FA-IMT同时检测其代谢指标。结果303例T2DM患者与93例健康对照基因型分布频率均符合Hardy-W e inberg氏平衡;健康对照组与T2DM组2组患者PAI-1基因型及等位基因频率比较差异无统计学意义(P>0.05);PAI-1各基因型人群的一般资料、代谢指标和FA-IMT等相比较差异无统计学意义(P>0.05)。结论PAI-1 4G/5G基因型多态性可能不是致T2DM的遗传易感因素,也可能不是致T2DM患者FA-IMT的遗传易感因素。Objective To investigate the relationship between the polymorphisms of plasminogen activator inhibitor-1 ( PAI-1 ) gene and the femoral artery intima-media thickness (FA-IMT) in patients with type 2 diabetes (T2DM). Methods The polymorphisms of PAI-1 (4G/5G) gene were determined by allele-specific polymerase chain reaction (AS-PCR) method and the FA-IMT was assessed using noninvasive high resolution B-mode uhrasonography in 303 T2DM patients and 93 controls in Hang population of Hunan province. The common data and metabolic index were compared among the groups with different genotypes of PAI-1. Results Frequencies of PAI-1 alleles and genotypes in 303 T2DM patients and 93 controls were all in accordance with the Hardy-Weinberg equilibrium, without significant differences between T2DM patients and controls ( P 〉0. 05 ). There were not significant differences in FA-IMT and all other data among different genotypes of PAI-1 ( P 〉0. 05). Conclusion The PAI-1 genotype perhaps may not be one of independent risk factors for both T2DM and FA-IMT in T2DM patients.
