摘 要:目的研究姜黄素对慢性心力衰竭大鼠左心室胶原网络重构的影响及其作用机制.方法采用缩窄腹主动脉的方法,将大鼠随机分成假手术组、手术对照组、卡托普利组和姜黄素组,姜黄素组又分为高、中、低三个剂量组,术后每周测定鼠尾动脉血压,分别在术后12,20wk观察心脏血流动力学变化,检测心肌胶原含量及心肌细胞TNFα的表达水平.结果12wk末,手术对照组左心室舒张压(LVEDP)、胶原容积百分比(CVF),肿瘤坏死因子(TNFα)的表达均明显强于其他各组,差别有统计学意义(P<0.05);20wk末,与手术对照组相比,姜黄素各剂量组和卡托普利组均能显著降低心衰大鼠的LVEDP,CVF,TNFα的表达(P<0.05),姜黄素高剂量组作用更强(P<0.01).不同时间段,手术对照组、卡托普利组和姜黄素各剂量组的前后差别均有统计学意义(P<0.05).结论姜黄素通过对TNFα的抑制作用及抗氧化作用等多种机制抑制胶原重构,在一定程度上改善心功能,延缓了心力衰竭的进程.AIM: To investigate the effects of curcumin on the remodeling of the left ventricular collagen network in rats with chronic heart failure (CHF) and its mechanism. METHODS: Pressure overloaded rat models were established by partial abdominal aorta ligation. The rats were divided into the shamoperated group, vehicle group, captopril group and curcumin group. Curcumin group was subdivided into the high-dose, medium-dose and low-dose groups. After operation, we detected rat caudal arterial blood pressure weekly and observed cardiac hemodynamics at the 12th and 20th week. The content of cardiac muscle collagen and the expression of TNFct were detected. RESULTS: At the end of the 12th week, the LVEDP, CVF-V, CVF-NV and TNFα in vehicle group were significantly increased compared with those in other groups ( P 〈 0. 05 ). Compared with that in vehicle group, the left ventricular function in curcumin group and captopril group were significantly ameliorated, as indicated by the decreased LVEDP, CVF-V, CVF-NV and TNFα (P 〈 0. 05 ), and the effect of high-dose curcumin group was better ( P 〈 0. 01 ). During different periods of CHF, there were significant differences in vehicle group, captopril group and curcumin groups ( P 〈 0. 05 ). CONCLUSION : It is the inhibitory effect of curcumin on TNFα and the oxidation that underlie the mechanism of curcumin in preventing collagen network remodeling and improving the cardiac function.
