摘  要:研究聚乳酸在骨修复过程中的降解特点以及在降解过程中对骨组织的影响,探讨它是否适于作为组织工程化骨的支架。在12只犬两侧下颌骨缺损区分别植入聚乳酸/骨髓基质干细胞/骨形成蛋白以及单纯PLA ,于术后2、4、8周观察聚乳酸形态以及植入部位的组织和细胞反应。结果表明,聚乳酸植入骨缺损后在表面和内部同时发生崩裂,至8周时大部分降解;降解过程中始终伴随着巨噬细胞活跃的吞噬活动;随着聚乳酸的降解,新生骨组织逐渐长入,骨缺损得到修复。聚乳酸在骨缺损内的降解时间超过8周;To investigate the degradation prodcess of polyactic acid(PLA) within bone reconstruction and its adverse effect on bone reconstruction. The bone defects of bilateral mandible of 12 dogs were implanted PIA/BMSc/rhBMP - 2 at one side, while PLA were implanted into the bone defect as control. The PIA and new regeneration bone were harvested at 2,4,8 weeks, and inspected by means of gross, histological observation and scanning electronic micmscope (SEM) examination. Results showed that the degradation occurred not only on the surface but also inside the PLA after implanted into the defects. PLA fragments still could be found at the end of this study. During the degradation of PLA, macrophages invaded into the PLA and exhibited active phagocytosis. There were mild degree infiltration of lymphocytes and fibroblast around the filaments during their degradation. With the degradation of PLA, the bone defects were reconstructed by the new bone. It takes more than 8 weeks that PLA degrades in bone defects completely. Mild nonspecific inflammation caused by implantation of PLA into bone defects, has no adverse effects on nerve.
