摘  要:目的:观察孕二烯酮对离体原代培养大鼠颗粒细胞分泌雌、孕激素功能及芳香化酶表达的抑制作用。方法:用活细胞计数法(CCK-8法)计算细胞存活率及半数抑制率(IC50)。加入卵泡刺激素和雄烯二酮刺激颗粒细胞分泌激素。用磁性均相酶联免疫法测定培养液中雌、孕激素含量。用大恒MIVNT显微图象分析软件分析免疫组化方法中芳香化酶表达。结果:孕二烯酮杀伤细胞的IC50为41μg/ml(95%可性限38.6-44.1μg/ml)。孕二烯酮5μg/ml,10μg/ml和20μg/ml在不影响活细胞数的情况下对颗粒细胞分泌雌激素的抑制率分别为4.5%,14.3%,38.7%;而其对分泌孕激素的抑制率分别为42.8%,53.1%和61.9%。免疫组化亦证明孕二烯酮能影响颗粒细胞中芳香化酶的表达。结论:孕二烯酮能直接抑制离体培养的大鼠颗粒细胞分泌雌、孕激素;且可能是通过抑制芳香化酶的活性而产生作用。Objective: To study the effect of Gestodene on granulosa cells' viability and steroidogenesis and aromatase function. Methods: The CCK-8 was used to measure cell survival rate and IC50. FSH and androstenedione were added to stimulate the steroid secretion, serono diagnostics was used to evaluate the estrigen and progesterone secretion respectively.We also carried out experiment on the expression of aromatase by Immunohistochemical staining. Rusules: IC50 of Gestodene to damage cells was 41 μg/ml (95% confidence interval 38.6±44.1 μg/ml).Gestodene 5 μg/ml, 10 μg/ml, and 20 μg/ml greatly inhibited the estrogen secretion from granulosa cells by 4.5%, 14.3% and 38.7%, respectively in the presence of 0.01 IU/ml FSH and 0.5 μmol androstenedione without affecting the number of viable cells. The secretion of progesteron markedly decreased by 42.8%, 53.1%, and 61.9% concurrently. Immunohistochemical studies showed that the expression of aromatase was reduced significantly in McCoy'5A medium of 40 μg/ml Gestodene obviously. Conclusion: Gestodene directly inhibited the steroidogenesis of granulosa cells,and may affect aromatase activity of granulose cells, too.
