摘  要:目的:采用时间分辨荧光免疫分析技术建立高灵敏度、高特异性的β-hCG的快速全自动超微量检测方法。方法:以β-hCG单克隆抗体812#包被板,双功能螯合剂异氰酸苄基二乙烯三胺四乙酸络合Eu3+标记β-hCG单克隆抗体827#,发光增强系统为以β-二酮体为主的增强液。采用结果:特异性试验显示,与AFP、LH均无交叉反应;平均回收率为96.5%;健全性佳;50例血清β-hCG的TRFIA检测与CLIA(BeckmanAcess)分析结果一致,相关系数为0.972。结论:β-hCG时间分辨荧光免疫分析法灵敏度高、稳定性好,具有很好的临床应用前景。平衡饱和法建立β-hCG时间分辨荧光免疫分析,数据采用Logit-Log法函数数据处理程序处理。Objective: To establish time-resolved fluroimmunoassay (TRFIA) of β-human chorionic gonadotropin (β-hCG) based on the direct sandwich technique. Methods: Monoclonal antibodies (McAb) against a specific antigenic site on β-hCG was 812# and the europium-labelled McAb against a different antigenic site on the β-hCG molecule was 827#. β-hCG in sera from patients and healthy controls were examined by β-hCG-TRFIA with auto DELFIA 1235 system. Results: The cross-reacting rates with AFP and LH were negative, respectively. The recovery rate is high in this experiment. We detected β-hCG in 50 sera by using TRFIA and CLIA methods, respectively. The correlation coefficient was 0.972. Conclusion: The newly developed β-hCG-TRFIA was of high sensitivity, specificity, stability.
