摘  要:目的探讨高血压大鼠心血管重构时,心肌细胞骨架蛋白α-微管蛋白(tubulin)和结蛋白(desmin)的变化。方法建立腹主动脉缩窄高血压大鼠模型,术后2周、4周用尾动脉测压法观察大鼠血压和心率的变化,术后11周用免疫组织化学和形态计量学方法,观察心血管形态改变、细胞骨架蛋白α-微管蛋白、结蛋白在心脏原位蛋白表达的变化。结果与假手术组比较,高血压大鼠2周和4周血压显著升高[(144·7±4·9比110·0±4·0)mm Hg,P<0·01;(151·4±14·9比114·4±5·5)mm Hg,P<0·01)、心血管重构,细胞骨架蛋白在心脏过表达和聚集、分布异常,而且阳性表达面积和阳性表达面积百分数增大,α-微管蛋白[(102927·8±20561·9比18304·4±9661·0)μm2,P<0·01]、结蛋白[(153271·7±15193·7比48686·1±14693·3)μm2,P<0·01]。结论细胞骨架蛋白α-微管蛋白、结蛋白表达上调,参与了高血压心血管重构时心肌细胞骨架重构。Objective To study the cytoskeletal remodeling in the eardiomyocytes in hypertensive rats. Methods Using a rat model of hypertension induced by abdominal aortic banding, we measured blood pressure with a tail cuff after 2 or 4 weeks of operation. The morphological change and the expression of the α-tubulin or desmin was studied after 11 weeks of operation. Results BP were increased (at 2nd wk.144.7±4.9 vs 110.0±4.0 mm Hg, P〈0.01; at 4th wk:151.4±14.9 vs 114.4±5.5 mm Hg, P 〈0.01) and compared with control rats, the eytoskeleton protein was overexpressed, and disorganized. The expression area and area percentage of α-tubulin area (102 927.8± 20 561.9 vs 18 304. 4± 9661.0 μm^2 , P〈0. 01) and desmin area (153 271.7± 15193.7 vs 48 686. 1 ± 14 693.3μm^2 , P〈0.01) were increased in the hypertensive rats. Conclusion The overexpression of α-tubulin and desmin were involved in the cardiovascular remodeling in the hypertensive rats.
