摘 要:目的建立测定人血浆中盐酸伪麻黄碱浓度的方法。方法血浆样品经混合碱液碱化、甲基叔丁基醚萃取、0.5%盐酸溶液酸化反提处理后,采用反相HPLC法测定。色谱柱:Kromasil C18柱(250mm×4.6mm,5μm),柱温30℃;流动相:0.01mol·L^-1磷酸盐缓冲液(pH6.5)-乙腈(62:38),流速1.0ml·min^-1;检测波长210nm。结果盐酸伪麻黄碱在0.015—1.60mg·L^-1范围内线性关系良好,回归方程为:Y=4631.8X+3.76(r=0.9998),该方法平均回收率为101.7%,日内RSD为3.41%-4.25%,日问RSD为6.47—9.66%。结论该法简便、灵敏、快速、稳定性好,可用于临床血药浓度检测及药代动力学研究。Aim To establish a senfive and specific method for determination of pseudoephcdrine hydrochloride in human plasma. Methods Pseudoephedrine was extracted from alkaline plasma with t-butyl methyl ether as the base form,and was back-extracted into 0.5% hydrochloride solution. The drug was determined by RP-HPLC. The LC-6A HPLC apparatus and Kromasil C18 column were used with mobile phase of 0.01 mol· L^-1 KH2 PO4 (pH6.5)-acetonltrile (62:38 ) ,The flow rate was 1.0ml · min^- 1 and the detection wavelength was 210 nm. Results The linear range of pseudoephedrine was 0.015 -1.60 mg·L^-1 ,(Y =4631.8X +3.76,r =0. 999 8) ,The average recovery of pseudoephedrine in human plasma was 101.7%. The within-day RSD were 3.41% -4.25% ,and the daytime RSD were 6. 47% - 9.66%. Conclusion The method was specific, sensitivity, simple and suitable for the determination of human plasma pseudoephedrine and pharmacokinefic researches.
