RP-HPLC测定奥昔布宁渗透泵控释片含量及有关物质的方法学研究

摘　　要：目的:建立 RP-HPLC 对奥昔布宁渗透泵控释片有关物质检查及含量的测定方法,为制剂的质量控制提供有效的分析方法。方法:采用 Kromasil C_8色谱柱(10μm,4.6mm×25cm),流动相:乙腈-pH 3.5三乙醇胺溶液(35:65),流速1mL·min^(-1);检测波长:200nm。结果:奥昔布宁与有关物质完全分离,奥昔布宁浓度在2.5～100μg·mL^(-1)内,与峰面积线性关系良好,相关系数0.9999,平均回收率为102.2%。HPLC 方法的系统适应性良好,回收率及精密度结果满足分析要求,有关物质检查符合规定。结论:本法简便、准确,专属性强,可用于奥昔布宁渗透泵控释片的质量控制。Objective：To develop an RP-HPLC method for the determination of oxybutynin（Oxb） and its related compounds in osmotic pump controlled - release tablets. Methods：Chromatography was carried out on a Kromasil C8 column（ 10 μm ,4. 6 mm × 25 cm）, using a mobile phase of acetonitrile -triethanolamine solution （pH 3.5 ） （35： 65） with a flow rate at 1.0 mL ·min ^- 1. The UV detective wavelength was 200 nm. Results： Linear range ： 2. 5 - 100. 0 μg · mL^-1, correlation coefficient：0. 9999, average recovery： 102. 2%. Chromatographic system suitability was good, average recovery and precision results were meet the needs of assay. Related substances test was up to the criterion. Conclusion： The method is convenient, accurate, specific and can be used in quality control of Oxb preparations.
