和络舒肝胶囊对日本血吸虫病肝纤维化的治疗作用研究

摘　　要：目的研究和络舒肝胶囊对小鼠日本血吸虫病肝纤维化的治疗作用及其机理,并与吡喹酮的作用对比。方法80只小鼠随机分为4组。其中3组小鼠感染日本血吸虫8周后,分别以和络舒肝胶囊0.1粒/只/d治疗8周、吡喹酮500mg.(kg.d)-1治疗2 d以及不作任何治疗(实验对照组),第4组小鼠作为正常对照组。应用免疫组化染色及HE染色法,观察和分析和络舒肝胶囊及吡喹酮治疗前后各组小鼠肝组织病理改变及血小板衍生生长因-BB(p latelet-derivedgrowth factor,PDGF-BB)和Ⅰ,Ⅲ型胶原的表达变化。结果和络舒肝胶囊治疗后肝纤维化组织病理损伤减轻,PDGF-BB和Ⅰ,Ⅲ型胶原含量均明显低于实验对照组(P<0.01)和吡喹酮组(P<0.01或P<0.05)。结论和络舒肝胶囊对小鼠日本血吸虫肝纤维化具有较好的治疗作用,其远期抗肝纤维化效果优于吡喹酮。Objective To study the therapeutical effects and mechanisms of Heluoshugan capsule on schistosome - infected mice with liver fibrosis. Compare the effects with praziquantel. Methods Eighty mice were divided into four groups ： three groups were infected with Schistosomajaponicum. After eight weeks, one group was treated with Heluoshugan capsule 0. lcapsule per mice evely day for 8 weeks, one group was treated with praziquantel 500mg/（ kg·d） for 2 days and the other was taken as experimental control without any treatment. The fourth group was taken as normal control, lmmunohistochemical technology and HE staining were applied to observe the content of hepatic PDGF - BB,type Ⅰ and type Ⅲ collagen in mice infected with S. japonieum before and after treatments. Results Heluoshugan capsule treatment significantly relieved pathologic damages in liver tissue with fibrosis, reduced the content of PDGF - BB, type Ⅰ and type Ⅲ collagen compared to the experimental control （P 〈 0.01 ） and the group treated with praziquantel （P 〈 0.01 or P 〈 0.05 ）. Conclusion Heluoshugan capsule has better therapeutical effects on Sehistosome - infected mice with liver fibrosis and the effects surpass praziquantel in the long run.
