摘 要:目的研究以3-巯丙基三乙氧基硅烷作为Fe3O4磁性粒子表面修饰剂,合成微米级磁响应性好且巯基含量较高的功能化磁性微粒,并对磁性微粒粒径以及表面巯基含量进行表征。方法激光粒度散射仪对功能化磁性微粒进行粒径分析,傅立叶变换红外光谱(FTIR)对巯基修饰前、后磁性微粒表面的功能基团进行表征,E llman方法对功能化磁性微粒表面巯基含量进行定量测定。结果氮气保护,乙醇与水体积比1∶1作为溶剂,500μL浓氨水为催化剂,加入300μL硅烷化试剂与170 mg磁性Fe3O4粒子充分反应,合成了平均粒径约为5μm的巯基化磁性微粒,所建立E llman方法对功能化磁性微粒表面巯基含量的测定结果为357μmol/g。结论功能化磁性微粒表面巯基的修饰及定量测定,为其在生物及医学领域的应用奠定了基础。Aim To synthesize Fe304 magnetic particles terminated with -SH group in different condition, the ratio of the Fe304 magnetic particles to 3-mercaptopropyhriethoxysilane, the catalyst and the protection atmosphere. The magnetic particles modified with-SH have small size, good magnetically responsive property and high amount of thiols. Methods Malvein particle size analyzer was used to analyze the size distribution of functional Fe204 particle, the thiol-functional Fe304 magnetic particle was characterized by the FTIR spectrum and Ellman's method was established and used for quantitatively analyzing the-SH group on particle surface respectively. Results Under the condition of N2,500μL concentration ammonia as the catalyst, 300μL of 3-m ercaptopropyltriethoxysilane and 170 mg of Fe2O4 were added to mixture solvent (ethanol: water = 1:1 ) and reacted for 6 h, the functional magnetic particles with 5μm average diameter was synthesized, the amount of thiol groups was 357μmoL/g determined by means of Ellman's method. Conclusion The modification and the quantitative methods for-SH groups on magnetic particle surface provide potential application for biological and medical areas in the near future.
