摘 要:目的建立家兔血浆和脑脊液中卡马西平(CBZ)及其活性代谢产物环氧化卡马西平(ECBZ)的高效液相色谱(HPLC)分析方法,并研究其药动学过程。方法选择奥卡西平为内标,采用Hypersil-ODS(250mm×4.6mm,5μm)色谱柱,以乙腈-水(63:37)为流动相,流速0.8mL·min^-1,柱温30℃,在紫外波长为210nm处检测。所得教据用3P87药动学程序处理,求出有关的药动学参数。结果CBZ和ECBZ在0.05~10.00mg·L^-1的浓度范围内线性关系良好(r=0.9999),平均回收率为99.6%~105.4%,日内、日间精密度RSD均〈7%。结论该方法简便、准确、灵敏度高,可用于CBZ和ECBZ的检测及药动学研究。Objective A HPLC method was established for the determination of carbamazepine (CBZ) and 10, 11-epoxide carbamazepine (ECBZ) in plasma and cerebrospinal fluid, and to investigate its pharmacokinetic characteristics in rabbits. Methods With oxcarbazepine as the internal standard, CBZ and ECBZ were detectded by UV detector at 210 nm with the Hypersil-ODS C18 column and acetonitrile-water(63 : 37) as the mobile phase at a flow rate of 0.8 mL·min^-1. The data obtained were treated with 3P87 pharmacokinetic program on computer. Results The linear range was 0.05 - 10.00 mg·min^-1 (r =0.999 9). The mean recoveries of the assay were 99.6% - 105.4%. Both the intra-day and inter-day Precisious(RSD) were less than 7%. Conclusion This method is simple, sensitive and accurate. It can be used for the detection and pharmacokinetic studies of CBZ and ECBZ in rabbits.
