摘 要:目的通过离体鼻黏膜的渗透实验筛选最佳促渗剂类型。方法采用透皮实验用扩散池装置用于透黏膜实验,HPLC法测定接受液中药物浓度,从而计算累计渗透药量。结果盐酸二氢埃托啡累计渗透药量与时间呈线性关系。与不含促渗剂的处方相比,促渗剂可显著提高药物的渗透速率,其中,尤以DM-β-CD促渗作用最强,其次为HP-β-CD和1,2-丙二醇,β-CD也有促渗作用。结论添加促渗剂可以提高盐酸二氢埃托啡的鼻黏膜渗透性,并且以DM-β-CD的促渗作用最强。Aim To pick out the best type of penetration enhancers through the test in vitro using excised sheep nasal mucosa. Methods Diffusion cells were adopted as apparatus for in vitro nasal mucosa. The concentration of dihydroetorphine hydrochloride was measured by HPLC and the cumulative penetration amount was calculated. Results The cumulative penetration amount of dihydroetorphine hydrochloride was linearly related to time. The penetration rate was significantly increased when penetration enhancers were used. Of these penetration enhancers, DM-β-CD was the best, and HP-β-CD and propylene glycol were the second best, followed by β-CD. Conclusion Penetration enhancers could enhance the penetration rate of dihydroetorphine hydrochloride, especially when DM-β-CD was added.
