摘 要:目的:观察金雀异黄素对去卵巢SD大鼠不同时期离体骨组织微损伤的影响及防治效果。方法:10月龄SD大鼠54只,随机分为3组各18只。将去卵巢的大鼠分为去卵巢组(A组)、金雀异黄素组(B组)各18只,另18只为对照组(C组)。B组术后每天给予金雀异黄素10μg/kg,共16周。于去卵巢后4、8和16周时处死3组大鼠各6只,对其第4离体椎骨进行疲劳损伤后观测椎骨变化。结果:疲劳损伤后每块骨组织磨片均可见微损伤,以微破裂最常见。3组之间椎体体积(V1)及微破裂平均长度(P4)比较差异均无显著性意义。骨小梁单位面积百分率在去卵巢4、8及16周时,A组低于B、C组(P<0.05);单位骨小梁面积百分率的微破裂长度与数目在16周时A组高于C组(P<0.05),在各个时期B、C组各项指标比较差异均无显著性意义。结论:大鼠去卵巢后的8周内尚不表现出骨结构性能的下降,金雀异黄素早期应用可使骨结构性能提高,长期应用能防止骨量丢失并改善骨结构。单位骨小梁面积百分率的微破裂长度和数目可反映药物对骨质疏松的影响,可作为骨结构性能的评价指标。Objective: To observe the effect of genistein on bone microdamage in ovarieetomized SD rats at different stages, evaluate the preventive and curative effectiveness of genistein. Methods: Fifty-four SD female rats (aged 10 months) were divided into 3 groups: group A, ovariectornlzed (OVX) t group B, genistein (10 btg/kg every day), and group C(control), At the 4th, 8th and 16th week after ovariectomy the rats were killed respectively. The 4th vertebrae were used for fatigue damage tGST by SL22000 bone fatigue experiment device. The parameters of microdamage were measured. Results: Mierodamage could he ohserved in each sample and microerack was the most usual damaged type. Significant difference in length and number of microcrack per percent oI trabecular area was observed (P〈0.05) between OVX group and genistein group at the 16th week, but not between OVX and control groups at the 4th and 8th week. Significant difference in length and number of microcrack per percent of trabecular area was not observed between genistein and control groups. Conclusion: Genistein could improve bone biological quality at the earlier stage and prevent it from deterioration later. The length and number of microcrack per percent of trabecular area could be used as bone hiomechanical marker to evaluate the effectiveness of antkosteoporostics.
